Summary: Ribonucleoside-diphosphate reductase subunit M2 (RRM2) is associated with prognosis and drug resistance in multiple myeloma. The inhibitor osalmid inhibits RRM2 translocation and regulates autophagosome synthesis and fusion with lysosomes, leading to autophagic cell death. Combination therapy with bafilomycin A1 further enhances autophagic cell death.

ANXA1 also counteracts the autophagic death of colon cancer cells, suggesting it could be a potential druggable target.

Summary: Myocardial infarction is a leading cause of death in patients with coronary heart disease worldwide. Myocardial ischemia/reperfusion injury (MIRI) results in more serious damage to the ischemic myocardium when the blood supply is restored. The mechanisms of MIRI involve oxidative stress, calcium overload, energy metabolism disorder, apoptosis, endoplasmic reticulum stress, autophagy, pyroptosis, necroptosis, and ferroptosis. Apoptosis and autophagy have received more attention, but necroptosis and ferroptosis also play important roles. However, the exact mechanism of MIRI remains unclear.
