Summary: Bone regeneration requires the interaction of multiple factors, including osteogenesis, osteolysis, angiogenesis, and an appropriate immune microenvironment. Recombinant human parathyroid hormone (PTH1-34) has been proven effective for clinical treatment of osteoporosis. However, its use in local bone repair is limited due to net bone resorption. This study proposes a strategy using multifunctional calcium phosphate (CaP) ceramics to modulate the effects of M1 macrophages and promote bone regeneration. The functionalization of the CaP ceramic scaffold with parathyroid hormone related peptide-1 (PTHrP-1) improves the osteogenic microenvironment, retains osteogenic and angiogenic properties, and achieves slow release of PTHrP-1. The results show that PTHrP-1-TBC is more suitable for bone regeneration than the unmodified peptide or the TBC scaffold alone.

Summary: The use of hydrogels containing PTH and nHAP can promote cranial bone regeneration by enhancing the expression of osteogenic-related proteins and facilitating osteogenic differentiation of BMSCs through the Notch signaling pathway. Implantation of the hydrogel into a rat cranial defect model showed efficient bone regeneration compared to using the hydrogel alone or nHAP alone, indicating a promising therapeutic effect of combining nHAP and PTH for bone regeneration.

Summary: This study investigated the relationship between 25(OH)D and 1,25(OH)2D with PTH. The results showed a negative correlation between all forms of PTH and 25(OH)D, while there was no significant correlation between 1,25(OH)2D and PTH. This finding suggests an inhibitory effect of 25(OH)D on the synthesis of all forms of PTH in the chief cells of the parathyroid gland.

Summary: In preterm newborns, secondary hyperparathyroidism is a frequently occurring but often unrecognized condition. Detecting this condition may be important in guiding nutritional treatment. This study found that cesarean section, oxygen use during resuscitation, invasive mechanical ventilation, and anemia requiring transfusion were independent risk factors for secondary hyperparathyroidism in preterm infants. Additionally, serum calcium, serum phosphate, vitamin D, and the calcium/creatinine ratio in urine were associated with secondary hyperparathyroidism. Furthermore, a predictive model using gestational age and calcium/creatinine ratio in urine demonstrated good accuracy.

Summary: This review highlights recent research on the regulatory functions of ubiquitin-specific peptidases (USPs) in osteoblasts and provides insight into the molecular mechanisms governing their actions. It also discusses the role of USP53 in osteoblasts and the complex layers of regulation exerted by USPs on osteoblast signaling. Further studies using knockout mouse models are necessary for a full understanding of the mechanisms underpinning USPs actions.

Summary: To systematically repair osteoporotic bone defects, a dynamically movable drug delivery system using monoporous microspheres (MPMs) encapsulating parathyroid hormone (PTH) was developed. The PTH loaded inside MPMs can be gradually released to improve drug concentration and reduce topical accumulation. In vivo evaluation showed that the implant of MPMs-PTH can stimulate regenerative repair of cranial defects and relieve osteoporosis throughout the body.

Summary: This study developed a composite hydrogel (PHC) by integrating short-chain chitosan and nanoparticulate hydroxyapatite into a covalent poly (ethylene glycol) network. The PHC hydrogel showed enhanced osteogenesis, high mechanical strength, excellent surface properties, and good biocompatibility. The hydrogel also had immunomodulatory effects, promoting M2 macrophage polarization and suppressing M1 macrophage polarization, while increasing osteogenic capacity and inhibiting osteoclast formation and resorption. Parathyroid hormone (PTH) could be effectively loaded and released in the PHC hydrogel (PTH@PHC), further improving osteogenic ability.

Summary: This study found that Insulin-like growth factor-binding protein 7 (IGFBP7) plays an important role in mediating the anabolic effects of parathyroid hormone (PTH). Intermittent PTH administration increased the expression of IGFBP7 in mesenchymal stem cells (MSCs) and pre-osteoblasts. Knockdown of IGFBP7 interrupted the anabolic effects of PTH, while supplementation with IGFBP7 protein enhanced the bone-forming efficacy of PTH. In a mouse model of fracture, administration of IGFBP7 along with PTH accelerated bone healing.
