Summary: The enzymatic activity of APOBEC3B has been implicated as a main cause of mutagenesis in head and neck squamous cell carcinoma. The expression of PKC alpha and NF-kappa B p65 has been linked to the upregulation of A3B. The study evaluated the expression of PKC alpha, NF-kappa B p65, and A3B in non-HPV related oral and oropharyngeal squamous cell carcinomas and found that A3B expression is associated with PKC alpha/NF-kappa B p65 expression, suggesting a potential role for the PKC/NF-kappa B signaling pathway in the development of oral and oropharyngeal cancer.

Summary: Oral cancer is a common type of cancer worldwide, and targeting cancer stem cells (CSC) may lead to promising results in terms of tumor relapse and metastasis. This study found that butylidenephthalide, a compound from Angelica sinensis, has anti-CSC effects in oral cancer. It inhibited the proliferation, migration, invasion, and colony formation abilities of CSCs, and also delayed tumor development.

Summary: By applying targeted spatial proteomic approaches, this study investigated the protein expression in primary and lymph node metastases and found that lymph node metastases had higher levels of proliferation, DNA repair, and apoptosis markers, while pro-apoptotic markers were enriched in the stroma of primary tumors. This study highlights the utility of spatial proteomics for understanding the tumor and stromal composition in locoregional metastasis.

Summary: Classical markers alone may not be enough to classify HPV-positive HNSCC patients, but when combined with serological markers, they are strong prognostic indicators in OPSCC patients. Detection of HPV16 E7 oncoprotein in serum at diagnosis is correlated with disease recurrence and patient overall survival, providing a useful tool for stratifying and monitoring HPV-positive HNSCC patients for worse prognosis.
