Summary: The recent classification of colon cancer into molecular subtypes has revealed that patients with poor prognosis have tumors with low levels of Wnt signaling, contrary to the previously understood role of overactive Wnt signaling in tumor progression. This study directly tests this assumption by reducing Wnt signaling in colon cancer cell lines and finds that decreased Wnt signaling leads to a more aggressive disease phenotype, with increased cell mobility and invasive behavior. RNA sequencing further reveals changes in gene programs related to cell migration and inflammation in the tumor microenvironment. These findings highlight the importance of carefully considering the impacts of targeting Wnt signaling in colon cancer therapy.

Summary: This study investigated the mechanism of MMP-9 secretion in airway epithelial cells infected with Mycoplasma pneumoniae. The results showed that M. pneumoniae infection induced the secretion of MMP-9 from bronchial epithelial cells and upregulated its enzymatic activity through the TLR2 and 6, MAPK/NF-kappa B/AP-1 signaling pathways, and histone acetylation. Additionally, M. pneumoniae infection decreased the expression of the tumor suppressor RECK, leading to increased MMP-9 activity.

Summary: The study found that chronic cold and capsaicin exposure further increased the deposition of collagen and elastin in colonic tissue. Increased expression levels of COL I and elastin mRNA and protein may contribute to enhanced ECM remodeling and stiffness variations of the colon tissue. Upregulated expression of LOXL2 and the physiological imbalance between MMP/TIMP activation and deactivation could potentially contribute to the progression of CRC induced by cold and capsaicin exposure.
