Summary: This study identified a rare PTGS1 variant associated with a bleeding phenotype and alterations in platelet reactivity and eicosanoid production in a large consanguineous family. The specific loss of platelet COX-1 activity was demonstrated for the first time in humans, providing important insights into platelet function and eicosanoid metabolism. Despite the absence of thromboxane A(2) formation by platelets, urinary thromboxane A(2) metabolites were in the normal range, suggesting the need for caution when using them as markers of in vivo platelet function.

Summary: Myocardial infarction (MI) is a significant cause of death in aged individuals, with females experiencing increased risk during the peri-menopausal period. Research shows that sEH levels are elevated in ischemic LV specimens, impacting oxylipid metabolism and mitochondrial enzymatic function. Targeting sEH with inhibitors or genetic deletion improves survival and cardiac function in female mice following MI.

Summary: Our study reveals that changes in protein content are associated with alterations in platelet structure and function as they age. Analysis suggests higher hemostasis in young platelets and increased apoptosis and senescence in old platelets. In-depth investigation shows reduced cytoskeletal protein, fewer mitochondria, and decreased calcium dynamics and granule secretion in aged platelets.

Summary: EETs have been shown to reduce macrophage activation and phagocytosis of Streptococcus pneumoniae by downregulating TLR2 and PGLYRP1 expression. Ephx2(-/-) mice had impaired lung bacterial clearance and reduced phagocytosis by macrophages.

Summary: The mammalian epoxide hydrolase (EPHX)3 is responsible for hydrolyzing epoxyalcohol derived triols in the 12R-lipoxygenase pathway and plays a crucial role in maintaining skin barrier integrity. This enzyme is essential for controlling metabolism flux through the alternative pathway and contributes to the transformation of naturally esterified epoxide substrates in other tissues.

Regulation of cardiovascular biology by microsomal epoxide hydrolase

Summary: Microsomal epoxide hydrolase (mEH) plays a crucial role in metabolizing compounds such as xenobiotics and pharmaceuticals by converting hydrophobic epoxides to hydrophilic diols. While mEH has been less studied in endogenous epoxy-fatty acid (EpFA) metabolism, recent findings suggest it may have a substantial impact on cardiovascular function through regulation of EpFA metabolism, particularly in the metabolism of epoxyeicosatrienoic acid (EET). Further research on the role of mEH in cardiovascular biology is warranted.
