Development of an innovative and sustainable one-step method for rapid plant DNA isolation for targeted PCR using magnetic ionic liquids

BackgroundNowadays, there is an increasing demand for fast and reliable plant biomolecular analyses. Conventional methods for the isolation of nucleic acids are time-consuming and require multiple and often non-automatable steps to remove cellular interferences, with consequence that sample preparation is the major bottleneck in the bioanalytical workflow. New opportunities have been created by the use of magnetic ionic liquids (MILs) thanks to their affinity for nucleic acids.ResultsIn the present study, a MIL-based magnet-assisted dispersive liquid-liquid microextraction (maDLLME) method was optimized for the extraction of genomic DNA from Arabidopsis thaliana (L.) Heynh leaves. MILs containing different metal centers were tested and the extraction method was optimized in terms of MIL volume and extraction time for purified DNA and crude lysates. The proposed approach yielded good extraction efficiency and is compatible with both quantitative analysis through fluorimetric-based detection and qualitative analysis as PCR amplification of multi and single locus genes. The protocol was successfully applied to a set of plant species and tissues.ConclusionsThe developed MIL-based maDLLME approach exhibits good enrichment of nucleic acids for extraction of template suitable for targeted PCR; it is very fast, sustainable and potentially automatable thereby representing a powerful tool for screening plants rapidly using DNA-based methods.

Isolation of DNA from plant tissues using a miniaturized matrix solid-phase dispersion approach featuring ionic liquid and magnetic ionic liquid solvents

Summary: A simple and rapid ionic liquid-based extraction method was developed for isolating genomic DNA from Arabidopsis thaliana plant tissue. The method involved using ionic liquid or magnetic ionic liquid directly applied to treated plant tissue to facilitate cell lysis and DNA extraction. The DNA-enriched extracts could be directly used in qPCR without needing additional purification steps and showed good preservation of plant DNA at room temperature.

This study synthesized hydrophobic [Ch+][Br-]-modified salts for the preparation of hydrophobic DESs. The [Ch+][Br-]-based HDESs were characterized and used as solvents for microextraction of phytochemicals in Cannabis sativa L. plant, showing good extraction performance for both hydrophilic and hydrophobic compounds.

Make the Quality Control of Essential Oils Greener: Fast Enantioselective GC-MS Analysis of Sweet and Bitter Orange as a Case Study

Assessing the environmental and overall performance of gas chromatographic analyses. Development of a comprehensive evaluation framework and application to routine chiral analyses of fragrances as a case study

Summary: Enantioselective gas chromatography is used in fragrance quality control laboratories to authenticate raw materials and detect fraud or adulteration of perfumes. This study developed a GC-MS method for chiral recognition of odorants and evaluated the environmental improvement of fast GC using AGREE. A multi-criteria approach based on the RGB model was developed, accurately accounting for energy and solvent consumption while maintaining analytical performance and laboratory productivity. The assessment showed that fast GC improves the greenness and productivity of a laboratory, lowering costs while maintaining adequate analytical performance, making it a sustainable approach.

Free Amino Acids and Biogenic Amines Profiling and Variation in Wild and Sub-Endemic Cardueae Species from Sardinia and Corse

Summary: This study aims to obtain, for the first time, the amino compounds profile of Cardueae plants using statistical analysis to detect similarities and differences among different samples. The results show that the DEEMM-derivatized amino compounds are qualitatively similar among the samples but can be discriminated at the genus level. This work provides more information on the phytochemical composition and potential health benefits of these little-studied plant species.
