Diagnostic Power of Cytokine M-CSF, Metalloproteinase 2 (MMP-2) and Tissue Inhibitor-2 (TIMP-2) in Cervical Cancer Patients Based on ROC Analysis

Macrophage colony-stimulating factor (M-CSF), matrix metalloproteinase-2 (MMP-2) and its specific tissue inhibitor (TIMP-2) may play an important role in the pathogenesis of cancer disease. We investigated the plasma levels and diagnostic power (ROC curve analysis) of M-CSF, MMP-2, TIMP-2 and tumor markers CA 125 and SCC-Ag in cervical cancer (CC) patients as compared to control group. The study included 89 patients with cervical cancer. The control group consisted of 50 healthy, untreated women. The plasma levels of M-CSF, MMP-2 and TIMP-2 were determined using ELISA, CA 125 and SCC-Ag - by CMIA method. The median levels of M-CSF, TIMP-2, SCC-Ag and CA 125 in the entire group of CC were significantly different than compared to the healthy women group. MMP-2 showed the highest value of sensitivity from all examined parameters (in stage I of CC - 93.10%, II - 82.76%, III and IV - 96.88%, total group - 92.05%). The highest specificity was obtained by M-CSF (86%). The area under the ROC curve (AUC) of M-CSF (0.8051) was the largest of all the tested parameters (even higher than commonly used tumor markers) in the group of cervical cancer. The combination of M-CSF, MMP-2 or TIMP-2 with SCC antigen resulted in an increase AUCs in all cases (0.8760;0.7880;0.8081;respectively). The findings of this study suggest the usefulness of all examined parameters in the diagnostics of CC patients. Out of the tested substances, M-CSF also appears to be the best candidate for cancer diagnostics in all stages of the disease, based on ROC analysis.

Circulating miRNAs were measured using the NanoString quantitative assay platform and validated with real-time polymerase chain reaction (RT-PCR). The results demonstrate the significance of miRNAs in the pathophysiology of GDM and their potential as predictive factors for early diagnosis.

Future Perspectives in Oxidative Stress in Trisomy 13 and 18 Evaluation

Summary: This review provides novel insights into the pathogenesis of trisomy 13 and 18, with a particular focus on oxidative stress. Evaluating oxidative stress is important for understanding disrupted metabolic pathways, mitochondrial dysfunction, and epigenetics. Additionally, unifying research protocols related to trisomy could have potential medical targets.

Metformin Intervention-A Panacea for Cancer Treatment?

Four microRNAs (miR-152-3p, miR-221-3p, miR-551b-3p, and miR-7-5p) were identified as potential diagnostic tools for PTC. This study highlights the potential of microRNAs as diagnostic biomarkers for PTC.

Utility of Matrix Metalloproteinases in the Diagnosis, Monitoring and Prognosis of Ovarian Cancer Patients

Summary: Ovarian cancer is a common gynecologic malignancy with a high mortality rate mainly due to its asymptomatic course. New diagnostic methods are being explored to enable earlier detection and better monitoring. Among these methods, extracellular matrix metalloproteinases (MMPs) are of particular interest. This article compiles the existing knowledge of MMPs in ovarian cancer patients and explores their potential diagnostic utility. Preliminary results indicate that MMPs may differ in tissue expression and blood and body fluid levels between ovarian cancer patients and healthy women. Moreover, the expression and concentration of individual MMPs have been shown to be correlated with cancer stage and disease severity.

Plasma Levels of Metalloproteinase 3 (MMP-3) and Metalloproteinase 7 (MMP-7) as New Candidates for Tumor Biomarkers in Diagnostic of Breast Cancer Patients

Summary: The aim of this study was to assess the plasma concentrations and diagnostic utility of MMP-3 and MMP-7 in early-stage breast cancer patients. The results showed that plasma levels of MMP-7 were significantly higher in Luminal A and Luminal B HER2-negative subtype breast cancer patients compared to the healthy group. MMP-7 demonstrated comparable or higher values of diagnostic sensitivity, specificity, positive and negative predictive values, and AUC compared to CA 15-3 or MMP-3. Therefore, combining MMP-7 with CA 15-3 may improve the diagnostic accuracy of breast cancer, particularly in Luminal B HER2-negative subtype patients, and MMP-7 could be a potential new tumor marker.

The Relationship between Oxidative Status and Radioiodine Treatment Qualification among Papillary Thyroid Cancer Patients

Summary: Markers of oxidative status, including total oxidative status (TOS), total antioxidant capacity (TAC), tumor protein 53 (p53), nuclear factor kappa B (NF-?B), forkhead box protein O1 (FOXO), and sirtuin 1 (SIRT1), play important roles in oxidative homeostasis and the progression of papillary thyroid cancer (PTC). Analyzing these markers can help determine eligibility for radioiodine (RAI) treatment. This study evaluated the relationship between oxidative status markers and qualification for RAI treatment in PTC patients.

CXC ELR-Positive Chemokines as Diagnostic and Prognostic Markers for Breast Cancer Patients

Summary: Currently, there is a constant search for tumor markers that can improve breast cancer diagnostics, especially in the early stages. Among the potential markers, chemokines have been found to be closely implicated in tumorigenesis and scientists have evaluated their usefulness in breast cancer diagnosis. This study summarizes the reports on the expression and peripheral blood concentration of ELR-positive CXC chemokines as potential markers and prognostic factors for breast cancer.

Clinical significance of oxidative stress markers as angioinvasion and metastasis indicators in papillary thyroid cancer

Summary: Angioinvasion remains an important prognostic factor in papillary thyroid cancer (PTC) patients. This study evaluated the utility of selected parameters in screening for angioinvasion and metastasis in PTC patients. The results showed that levels of serum antioxidant capacity (TAC) and sirtuin 3 (SIRT3) were decreased, while levels of DNA/RNA oxidative stress damage products (DNA/RNA OSDP) and malondialdehyde (MDA) were increased in PTC patients with angioinvasion and metastasis. The highest screening accuracy for detecting angioinvasion and metastasis in PTC patients was achieved using DNA/RNA OSDP, SIRT3, and TAC.

Identification and subsequent validation of transcriptomic signature associated with metabolic status in endometrial cancer

Summary: Aberrant metabolism has been identified as a main driver of cancer. In this study, transcriptomic profiles of metabolism-related pathways in endometrial cancer (EC) were established using NanoString nCounter Technology. 11 deregulated genes associated with 'central carbon metabolism in cancer' were identified, and drug repurposing validated several drug candidates. These findings have significant implications for personalized therapeutic strategies in EC.
