Formononetin attenuates kidney damage in type 2 diabetic rats

Aim: Diabetic nephropathy is the commonly developed complication of vasculature in type 2 diabetic patients. Chronic hyperglycemia leads to nephropathy in diabetics because of the formation of excessive reactive oxygen species and advanced glycation end products which is reflected in the form of glomerulosclerosis, tubular atrophy and interstitial fibrosis. As per the various reports reduction in SIRT1 expression in kidney tissue is key factor in the development of nephropathy in diabetes because its reduction in tissue is linked with excessive formation of ROS. Formononetin is a polyphenolic compound reported for its effect on SIRT1 and ROS. Main methods: Type 2 diabetes was induced in rats by diet modification using high fat diet for fifteen days prior to streptozotocin regimen (35 mg/kg, i.p.). Treatment of formononetin was started after confirmation of diabetes and continued for 16 weeks. Formononetin was administered orally to the diabetic animals at the dose of 10. 20 and 40 mg/kg. Key findings: Formononetin treatment for 16 week was able to control hyperglycemia and insulin resistance in diabetic animals. It has also been reduced triglyceride and cholesterol in blood. Formononetin treatment reduced blood concentration of creatinine, blood urea nitrogen and increased albumin concentration. Formononetin treatment also enhanced creatinine clearance in diabetic animals. Oxidative stress burden was also reduced significantly after formononetin treatment along with increased SIRT1 expression in kidney tissues of diabetic animals. Significance: Formononetin is a potential molecule which increases the expression of SIRT1 in kidney tissue of diabetic. Thus formononetin is an effective molecule to control nephropathy in type 2 diabetes mellitus.

Effect of Costus pictus per se and in combination with Metformin and Enalapril in streptozotocin induced diabetic nephropathy in rats

Summary: This study investigated the effects of a combination therapy of metformin, enalapril, and methanolic extract of Costus pictus (MECP) on diabetic nephropathy in rats. The combination therapy showed beneficial effects in terms of increasing body weight, reducing blood glucose levels, and improving kidney hypertrophy index. It also normalized the altered levels of serum and urine parameters. Histopathological evaluation revealed a reduction in tubular vacuolar degeneration.

Neuroprotective Effect of Quercetin and Memantine against AlCl3-Induced Neurotoxicity in Albino Wistar Rats

Summary: Recent evidences suggest a rise in global cases of dementia, with Alzheimer's disease being the leading cause. Quercetin, a flavonoid, has shown potential therapeutic benefits including neuroprotective effects. This study evaluates the neuroprotective effect of orally administered quercetin with memantine in albino Wistar rats after induction of neurotoxicity. The results demonstrate that the treatment of quercetin with memantine improves behavioral parameters, reduces oxidative stress, inhibits amyloid-beta plaque formation, and enhances the expression of brain-derived neurotrophic factor (BDNF).

Effects of baicalein with memantine on aluminium chloride-induced neurotoxicity in Wistar rats

Summary: Alzheimer's disease is a common form of dementia, and there are few approved medications for its treatment. Baicalein, a flavonoid, has shown neuroprotective effects, while memantine is an important drug for Alzheimer's treatment. This study investigated the combination of baicalein and memantine in treating aluminum chloride-induced neurotoxicity in rats. The combination treatment showed significant improvement in behavior, reduced oxidative stress and amyloid plaque formation, and increased BDNF expression. These findings suggest that baicalein and memantine may slow neurodegeneration progression.

Summary: Alzheimer's disease is a neurodegenerative condition characterized by progressive loss of memory and cognitive dysfunction, particularly in older individuals. The prevalence of the disease has increased due to the global aging population. Current treatments for Alzheimer's disease have limited efficacy and adverse effects. Recent research suggests that natural products, such as baicalein, could be potential treatment options for Alzheimer's disease. Baicalein, a flavonoid from the flavone subclass, has shown neuroprotective properties through its antioxidant, anti-inflammatory, AChE enzyme inhibitory, and anti-amyloid protein aggregation activities.
