N-acetylcysteine and alpha-lipoic acid improve antioxidant defenses and decrease oxidative stress, inflammation and serum lipid levels in ovariectomized rats via estrogen-independent mechanisms

Sexual hormone deficiency has been associated with metabolic changes, oxidative stress and subclinical inflammation in postmenopausal women. Hormone replacement therapies are effective in many instances, even though some patients either do not respond or are not eligible. The aim of this study was to evaluate the impact of short- (15 days) versus long-term (60 days) sexual hormone depletion and whether antioxidant supplementation with N-acetylcysteine (NAC) and alpha-lipoic acid (LA) improves oxidative stress, metabolic, and inflammatory parameters in ovariectomized (OVX) rats. Short-term OVX rapidly depleted circulating estrogen, causing uterine atrophy and body weight gain without affecting oxidative damage, inflammatory and lipid metabolism markers. In contrast, long-term OVX augmented oxidative damage in serum and peripheral tissues as well as increased serum total cholesterol, TNF-alpha and IL6 levels. Triglycerides, glucose and HDL cholesterol were not altered. Long-term OVX-induced oxidative stress was associated with depletion of GSH and total non-enzymatic antioxidants as well as decreased activity of Glutathione Peroxidase (GPx) and Glutathione Reductase (GR), but not Superoxide Dismutase (SOD) and Catalase (CAT). NAC and LA supplementation prevented GSH and total non-enzymatic antioxidants depletion as well as restored GPx and GR activities, TNF-alpha, IL6 and cholesterol in OVX rats. NAC and LA effects appear to be independent on NRF2 activation and estrogen-like activity, since NAC/LA did not promote NRF2 activation and were not able to emulate estrogen effects in OVX rats and estrogen-receptor-positive cells. The herein presented data suggest that NAC and LA may improve some deleterious effects of sexual hormone depletion via estrogen-independent mechanisms. (C) 2019 Elsevier Inc. All rights reserved.

Prevention of hospital pathogen biofilm formation by antimicrobial peptide KWI18

Perinatal exposure to a glyphosate pesticide formulation induces offspring liver damage

Summary: The present study investigated the effects of perinatal exposure to glyphosate-based herbicide (GBH) in offspring's liver. The results showed that perinatal exposure to GBH increased Ca2+ influx, oxidative damage and inflammation in the liver of offspring. Additionally, the study found that GBH exposure led to increased levels of iron in the liver, blood and bone marrow, which might contribute to iron-driven hepatotoxicity. These findings indicate the potential harmful effects of GBH on liver function in offspring.

Omics-based identification of an NRF2-related auranofin resistance signature in cancer: Insights into drug repurposing

In vivo exposure to pyriproxyfen causes ovarian oxidative stress and inhibits follicle maturation in zebrafish

Summary: We studied the effects of pyriproxyfen on zebrafish ovarian morphology and oxidative stress. Exposure to pyriproxyfen resulted in increased generation of reactive oxygen species and decreased glutathione content in the ovaries. Additionally, enzyme activities related to antioxidative defense mechanisms were altered. The histological analysis showed disrupted follicular maturation and various abnormalities in the ovary. These findings suggest that pyriproxyfen acts as an endocrine disruptor and can impair fish reproduction.

Selenylated Imidazo [1,2-a]pyridine Induces Apoptosis and Oxidative Stress in 2D and 3D Models of Colon Cancer Cells

Summary: Incidence rates of colon cancer are increasing annually due to genetic and epigenetic alterations that lead to drug resistance. Recent studies have found that novel synthetic selenium compounds, such as MRK-107, are more effective and less toxic than conventional drugs, exhibiting biocompatibility and pro-oxidant effects on tumor cells. MRK-107 inhibits cell proliferation, prevents cell regeneration and metastatic transition, and induces apoptosis in colon cancer cells through oxidative stress and activation of apoptotic pathways.

Could pesticide exposure be implicated in the high incidence rates of depression, anxiety and suicide in farmers? A systematic review

Summary: Pesticide exposure and poisoning can increase the risk of mental health problems and suicidal tendencies. A systematic review found that farmers exposed to pesticides had a higher prevalence of depressive disorders, and previous pesticide poisoning increased the risk of depression. Financial difficulties and poor health were also linked to depression in this population. Additionally, suicide rates were higher in areas with intensive pesticide consumption, highlighting the need for greater attention to the mental health of farmers and further research on occupational exposure to pesticides.

Regulatory effect of Epigallocatechin-3-O -gallate on circular RNA expression in mouse liver

Summary: This study investigates the impact of maternal vitamin D levels during pregnancy on infantile eczema. The findings suggest that lower prenatal maternal vitamin D levels increase the risk of eczema in infants aged 0-1 year. Furthermore, the study indicates that infants with eczema have lower expression of the Foxp3 gene in cord blood and decreased placental FOXP3 protein expression. These results suggest that vitamin D may affect the development of infantile eczema through the regulation of gene and protein expression in cord blood and placenta.

The study reveals that FTO is associated with hypothalamic leptin resistance and provides new insight into its role in obesity.

Regulation of the Notch signaling pathway by natural products for cancer therapy
