Summary: Bidirectional dialogue between cellular and non-cellular components of the tumor microenvironment (TME) drives cancer survival. The interplay between the matrix molecule tenascin-C (TNC) and chemokine CCL2 in HER2+ breast cancer was studied, revealing their distinct roles in subverting anti-tumoral immunity. TNC supported sustained CCL2 synthesis and facilitated an immune suppressive macrophage phenotype, while CCL2 did not impact macrophage survival/activation.

Summary: Breast cancer is a common cancer worldwide, and metastasis is the main cause of cancer-related death. Human monocyte chemoattractant protein-1 (MCP-1/CCL2) was identified as a chemotactic factor for monocytes, and its role in cancer development and progression was investigated. Studies showed positive correlations between MCP-1 production in tumors and TAM infiltration, as well as cancer progression. MCP-1 was found to promote breast cancer metastasis to the lung and brain, but not bone. The mechanisms of MCP-1 production in the breast cancer microenvironment were also reported.

Summary: CCR2 is predominantly expressed by monocytes/macrophages with proinflammatory functions, and targeting CCR2 has been investigated as a strategy to modify the tumor microenvironment and enhance anti-tumor immunity. In addition to blocking chemotaxis of suppressive myeloid cells, CCR2 signaling has underappreciated effects on myeloid cell survival and function polarization. T cells also express CCR2, and targeting CCR2 is shown to favor the anti-tumor arm of immune responses by affecting tissue homing of Treg cells and enhancing Treg functional potency.

Summary: Polyinosine-polycytidylic acid (pIC) inhibits the growth of pancreatic ductal adenocarcinoma (PDAC) by activating immune cells and inducing apoptosis. Delivery of pIC into PDAC cells inhibits tumor growth, promotes toxic autophagy, and induces apoptosis in animal models and in vitro. In vivo experiments show that pIC has strong antitumor immune activity and high effectiveness against PDAC, especially when used in combination with standard of care treatment.

Summary: A sandwich enzyme-linked dendrimer-aptamer assay (ELDAA) was developed to improve the detection performances of a traditional enzyme-linked immunosorbent assay (ELISA) using glass-bottom 96-micro-well plates. Poly(amidoamine) (PAMAM) dendrimers were immobilized onto the microwell surfaces and aptamers against human platelet-derived growth factor BB (PDGF-BB) were conjugated onto the surfaces via the immobilized PAMAM dendrimer templates. The ELDAA demonstrated a broader linear detection range and a lower limit of detection compared to the standard ELISA.

Summary: Chronic inflammation can lead to autoimmune diseases like rheumatoid arthritis and atherosclerosis. CCL2, also known as MCP-1, plays a crucial role in the progression of these diseases by attracting monocytes to the site of injury and promoting inflammation. In this study, computational modeling techniques were used to design high-affinity peptide binders for CCL2, with the aim of preventing its binding to CCR2 and reducing inflammation. Further analysis and experiments are needed to validate the accuracy of this computational approach in designing CCL2 cytokine binders.
