Taurine-upregulated gene 1 (TUG1) is a 7.1kb long noncoding RNA (lncRNA) first recognized in 2005 as an important element for retinal development in rodents. Subsequently, this lncRNA has been shown to participate in oncogenic processes through alteration in chromatin structure, sponging microRNAs, and affecting the expression of some cancer-related pathways. While most of the studies have revealed an oncogenic role for this lncRNA, some reports have shown downregulation of TUG1 in lung cancer samples compared with noncancerous samples. In triple negative breast cancer samples, the expression of this lncRNA has been decreased. Besides, its expression has been higher in HER2-enriched and basal-like subtypes compared with luminal A. In the current review, we discuss the latest literature about the expression pattern and functional roles of TUG1 in diverse cancer types. In addition, its role in epithelial-mesenchymal transition and activation of Wnt/beta-catenin pathway in human malignancies will be explored.

Emerging role of extracellular vesicles in the pathogenesis of glioblastoma

Maryam Khayamzadeh, Vahid Niazi, Bashdar Mahmud Hussen, Mohammad Taheri, Soudeh Ghafouri-Fard, Mohammad Samadian

Summary: Brain tumors, particularly glioblastoma, are aggressive and often fatal due to inadequate treatment and late detection. Recent studies have found that glioblastoma cells produce extracellular vesicles containing miRNAs and other components that enhance their malignant behavior and impact neighboring cells. Understanding the molecular composition of these extracellular vesicles is crucial in developing effective therapies against this type of tumor.

Expression of Treg-associated lncRNAs in breast cancer

Lina Moallemi-Rad, Amin Ghorbani, Maryam Dadyar, Bashdar Mahmud Hussen, Mohammed Fatih Rasul, Solat Eslami, Mohammad Taheri, Elena Jamali, Soudeh Ghafouri-Fard

Summary: This study evaluated the expression of five Treg-related long non-coding RNAs in breast cancer tissues. The results showed significantly higher expression levels of RMRP, TH2-LCR, MAFTRR, and GATA3-AS1 in breast cancer samples compared to non-tumoral tissues. Positive associations were found between the expression levels of RMRP and MAFTRR genes in tumor tissues and certain tumor characteristics. Additionally, differences in FLICR expression were observed among tumors with different levels of the HER2/neu receptor. These findings suggest that Treg-associated lncRNAs may contribute to the pathogenesis of breast cancer.

Summary: Recent decades have witnessed significant progress in understanding the role of different long non-coding RNAs (lncRNAs) in human diseases. ADAMTS9-AS2 is a notable lncRNA with diverse roles in various disorders. While it is generally recognized as a tumor suppressor lncRNA in different cancers, it has been found to be up-regulated in tongue squamous cell carcinoma, salivary adenoid cystic carcinoma, and glioblastoma. Additionally, ADAMTS9-AS2 may play a role in the pathoetiology of pulpitis, acute ischemic stroke, type 2 diabetes, and its complications. This review aims to summarize the role of ADAMTS9-AS2 in different diseases, particularly focusing on its diagnostic and prognostic values.

Summary: In order to control or stop the burden of cancer, various methods have been applied. However, these treatments often fail due to drug resistance or cancer recurrence. Combining the modulation of non-coding RNA (ncRNA) expression with other treatments can increase treatment sensitivity, but there are still challenges to overcome. Gathering information in this field is essential for finding more efficient cancer cures.
