Summary: Our study demonstrates that GQAs can be used as safe and highly effective phytochemicals against multidrug-resistant (MDR) Candida albicans. GQAs exhibited strong antifungal activity by interfering with fungal growth, disrupting fungal membrane integrity, and inducing apoptosis. Both in vitro and in vivo models confirmed the potential therapeutic use of GQAs for the treatment of vaginal candidiasis.

Summary: Mycoses are responsible for millions of infections worldwide each year, with invasive candidiasis being the most common and having high morbidity and mortality. The incidence of other Candida species, such as Candida parapsilosis, Candida glabrata, and Candida auris, is increasing. These pathogens often show reduced susceptibility to commonly used antifungal drugs, and the emergence of multi-drug-resistant strains continues to rise. Therefore, the search for new molecules that target these pathogens in a different way is urgent. Antimicrobial peptides, found in nature, have advantages over traditional antifungal agents and are considered a promising option for treating a wide range of microbial infections.

Summary: Starch materials tend to lose their initial characteristics, such as retrogradation degeneration, high hygroscopicity, and promoting microbial growth, which reduces the shelf life of packaging. Therefore, the incorporation of nanoparticulated CuO was evaluated for enhancing performance. CuO, as a semiconductor, improves optical, mechanical, and antimicrobial properties, making it suitable for improving starch films. By using the hot injection precipitation method, CuO nanoparticles in the nanometric scale were rapidly obtained. The resulting CuO-reinforced starch films exhibited improved tensile strength, barrier properties against radiation, and fungistatic effect against the Alternaria alternata fungus.

Summary: This study investigated the antifungal susceptibility and molecular epidemiology of vaginal and non-vaginal Candida glabrata isolates. The results showed high resistance rates to fluconazole, itraconazole, and voriconazole, while all isolates remained susceptible to amphotericin B. Genotyping revealed partially shared genotypes between vaginal and non-vaginal samples, with additional genotypes found in vaginal samples. The majority of resistant strains correlated with the predominant genotype.

Summary: This study reports the preparation and characterization of the amorphous form of Nifurtimox, a drug for Chagas disease treatment. The amorphous material is found to be sensitive to temperature, pressure, and mechanical stress, but it can remain stable for more than one month in a polymeric amorphous solid dispersion with PEG 4000. The study also discloses an approach for monitoring the conversion of the amorphous phase to its crystalline counterpart under thermal stress using chemometric analysis of mid-infrared spectra at different temperatures.

Summary: Tioconazole was prepared as a hydrochloride salt to improve its solubility and bioavailability. Single crystals were grown from an aqueous hydrochloric acid solution and their structure was determined. The salt exhibited much faster dissolution rate compared to tioconazole, indicating its potential as a replacement for the free base in certain applications.

Summary: Oral candidiasis is an opportunistic infection mainly affecting individuals with weakened immune systems. This study aimed to develop buccal films loaded with lidocaine and miconazole nitrate to topically treat this pathology. The films showed adequate adhesiveness and thickness and provided a fast release of lidocaine and a controlled release of miconazole nitrate with antifungal activity for 24 hours, reducing the need for daily applications compared to conventional treatments.

Summary: The development of age-appropriate formulations in the pharmaceutical industry is challenging, especially for pediatric and geriatric patients who may have difficulty swallowing oral medications. This study aimed to obtain a new dosage form of Carvedilol suitable for pediatric and geriatric patients. The solubility of Carvedilol was improved by analyzing and optimizing various factors, and the physical stability of the formulations was enhanced by adding HPMC. Microbiological tests were conducted to assess the stability of the formulations.

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
