Exosomes derived from human dermal papilla cells promote hair growth in cultured human hair follicles and augment the hair-inductive capacity of cultured dermal papilla spheres

Dermal papillae (DP) play key roles in hair growth and regeneration by regulating follicular cell activity. Owing to the established roles of exosomes (Exos) in the regulation of cell functions, we investigated whether DP-derived Exos, especially those from three-dimensional (3D)-cultured DP cells, affect hair growth, cycling and regeneration. Exos derived from 3D DP (3D DP-Exos) promoted the proliferation of DP cells and outer root sheath (ORS) cells and increased the expression of growth factors (IGF-1, KGF and HGF) in DP cells. 3D DP-Exo treatment also increased hair shaft elongation in cultured human hair follicles. In addition, local injections of 3D DP-Exos induced anagen from telogen and also prolonged anagen in mice. Moreover, Exo treatment in human DP spheres augmented hair follicle neogenesis when the DP spheres were implanted with mouse epidermal cells. Similar results were obtained using Exos derived from 2D-cultured DP cells (2D DP-Exo). Collectively, our data strongly suggest that Exos derived from DP cells promote hair growth and hair regeneration by regulating the activity of follicular dermal and epidermal cells; accordingly, these findings have implications for the development of therapeutic strategies for hair loss.

Targeting GLI1 Transcription Factor for Restoring Iodine Avidity with Redifferentiation in Radioactive-Iodine Refractory Thyroid Cancers

Summary: In this study, the researchers found that reversing iodine non-avid thyroid cancers to iodine avid cancers can make the cancers sensitive to radioactive-iodine therapy again. They demonstrated that GLI1 knockdown increased expression of thyroid-specific proteins, restoring iodine avidity and improving the efficacy of radioactive-iodine therapy. Targeting GLI1 could be a potential strategy for redifferentiation in dedifferentiated thyroid cancers.

Growth factor-rich and stem cell-rich treatments have shown potential, but standardization is lacking. Further understanding and standardization of regenerative cellular therapies for hair restoration are needed.

Summary: Various forms of platelet concentrates have evolved over the past few decades, with newer generation products such as leukocyte-platelet-rich fibrin (L-PRF) and advanced platelet-rich fibrin (A-PRF) showing superior biological properties in musculoskeletal regeneration compared to the first-generation concentrates. These newer platelet concentrates have a complete matrix of physiological fibrin and facilitate intercellular signaling and migration, promoting angiogenic, chondrogenic, and osteogenic activities. A-PRF, with higher leukocyte inclusion, also possesses antimicrobial activity and can replicate various physiological and immunological factors of wound healing. Its applications have expanded to include the management of musculoskeletal conditions, reconstructive cosmetic surgery, wound healing in diabetic patients, and maxillofacial surgeries.

The secretomes of 3D-MSCs promote proliferation, migration, and tube formation of ECs, and show enhanced wound healing in a burn injury mouse model.
