Extracellular Vesicles Long RNA Sequencing Reveals Abundant mRNA, circRNA, and lncRNA in Human Blood as Potential Biomarkers for Cancer Diagnosis

BACKGROUND: Extracellular vesicles (EVs) contain a rich cargo of different RNA species with specialized functions and clinical applications. However, the landscape and characteristics of extracellular vesicle longRNA(exLR) in human blood remain largely unknown. METHODS: We presented an optimized strategy for exLR sequencing (exLR-seq) of human plasma. The sample cohort included 159 healthy individuals, 150 patients with cancer (5 cancer types), and 43 patients with other diseases. Bioinformatics approaches were used to analyze the distribution and features of exLRs. Support vector machine algorithm was performed to construct the diagnosis classifier, and diagnostic efficiency was evaluated by ROC analysis. RESULTS: More than 10000 exLRs, including mRNA, circRNA, and lncRNA, were reliably detected in each exLR-seq sample from 1-2 mL of plasma. We observed that blood EVs contain a substantial fraction of intact mRNAs and a large number of assembling spliced junctions; circRNA was also enriched in blood EVs. Interestingly, blood exLRs reflected their tissue origins and the relative fractions of different immune cell types. Additionally, the exLR profile could distinguish patients with cancer from healthy individuals. We further showed that 8 exLRs can serve as biomarkers for hepatocellular carcinoma (HCC) diagnosis with high diagnostic efficiency in training [area under the curve (AUC) = 0.9527; 95% CI, 0.9170-0.9883], validation cohort (AUC = 0.9825; 95% CI, 0.9606-1), and testing cohort (AUC = 0.9627; 95% CI, 0.9263-0.9991). CONCLUSIONS: In summary, this study revealed abundant exLRs in human plasma and identified diverse specific markers potentially useful for cancer diagnosis. Colorectal Cancer-Derived Small Extracellular Vesicles Promote Tumor Immune Evasion by Upregulating PD-L1 Expression in Tumor-Associated Macrophages

Summary: This study reveals that colorectal cancer cells can induce macrophage polarization and PD-L1 expression through the release of small extracellular vesicles (sEVs), leading to suppressed CD8(+) T cell activity and increased tumor growth. The miR-21-5p and miR-200a derived from sEVs play key regulatory roles in this process. Inhibiting the secretion of specific sEV-miRNAs from CRC and targeting PD-L1 in TAMs may serve as novel methods for CRC treatment.

A pathway-guided strategy identifies a metabolic signature for prognosis prediction and precision therapy for hepatocellular carcinoma

Summary: The study presents a computational framework based on metabolic pathways to establish a metabolic signature for predicting the prognosis of hepatocellular carcinoma (HCC). The authors built an eight-gene risk score from 20 downregulated metabolic pathways in HCC and successfully validated the model in multiple independent cohorts. Additionally, three subtypes of HCC with distinct clinical outcomes were identified, and strong negative associations with other molecular features were found. The study also demonstrated the sensitivity of high-risk score subtypes to three potential therapeutic agents.

circTMEM181 upregulates ARHGAP29 to inhibit hepatocellular carcinoma migration and invasion by sponging miR-519a-5p

Summary: In this study, a novel circRNA, circTMEM181, was found to be downregulated in hepatocellular carcinoma tissues. Decreased expression of circTMEM181 was associated with shorter overall survival of HCC patients. Overexpression of circTMEM181 reduced the migration and invasion abilities of HCC cells, while knockdown of circTMEM181 increased cell motility.

Nuclear complement C3b promotes paclitaxel resistance by assembling the SIN3A/HDAC1/2 complex in non-small cell lung cancer

Summary: In addition to its classical role in innate immunity, intracellular complement components, particularly C3, play essential roles in immune defense, T cell homeostasis, and tumor cell proliferation and metastasis. This study demonstrates that C3 is significantly upregulated in PTX-resistant NSCLC cells and silencing C3 sensitizes resistant cells to PTX therapy. C3b translocates into the nucleus and interacts with the SIN3A complex to repress the expression of GADD45A, resulting in decreased PTX-induced apoptosis. These findings highlight the oncogenic properties of C3 and provide a potential therapeutic strategy to overcome PTX resistance.

Summary: ALDOA promotes liver cancer growth and metastasis by accelerating mRNA translation independent of its catalytic activity. It interacts with IGF2BP1 to facilitate its binding to m(6)A-modified eIF4G mRNA, increasing eIF4G protein levels and enhancing overall protein biosynthesis. Targeting ALDOA with GalNAc-conjugated siRNA effectively slows the tumor growth of orthotopic xenografts.

Summary: Sufficient evidence has linked many different types of cancers and T2D through shared risk factors; however, the underlying mechanisms are not fully understood. Increased levels of a-hydroxybutyrate (a-HB), a metabolite found in diabetes and cancer, including colorectal cancer (CRC), triggers the translocation of lactate dehydrogenase A (LDHA) to the nucleus. This process leads to an increase in TNF-a production, impaired insulin secretion, and exacerbation of CRC and type 2 diabetes. Metformin can interrupt this process by downregulating LDHA expression, thus highlighting its potential in preventing cancer and diabetes.
