Summary: Evidence suggests that non-coding RNAs, controlled by competing endogenous RNAs (ceRNAs), play a role in glioblastoma multiforme (GBM), although their regulatory mechanisms are still poorly understood. This study extensively analyzed the changes in circRNA, lncRNA, miRNA, and mRNA expression in GBM patients. The researchers identified differentially expressed genes (DEGs), lncRNAs (DELs), miRNAs (DEMs), and circRNAs (DECs) in GBM, and found that CEACAM5, CXCL17, FAM83A, TMPRSS4, and GGPRC5A were hub genes enriched in modules. Furthermore, a ceRNA network involving circRNA, lncRNA, miRNA, and mRNA was constructed, suggesting potential therapeutic targets for GBM.

Summary: In this study, the role of lncRNAs and circRNAs in the pathogenesis of HCC was investigated using expression profiles from a database. A lncRNA-mRNA network was established based on their correlation, and a circRNA/lncRNA-miRNA-mRNA co-expression network was constructed. Among the differentially expressed genes in this network, 55 miRNAs were found to be closely related to the prognosis of HCC. These findings provide new insights for further research and treatment of HCC.

Summary: This study focused on the transcriptomic profile of different subtypes of hUMSC-EVs using nanopore sequencing. The results showed abundant transcript variants in EVs, with a higher proportion of intact or near-complete transcripts in exosomes compared to microvesicles. Although the composition of RNA biotypes was similar between different EV subtypes, the distribution of transcripts and genes revealed heterogeneity between exosomes and microvesicles. Additionally, different expression patterns and pathway enrichments were observed, suggesting distinct functional tendencies of EV subtypes.

Summary: The analysis of extracellular vesicles (EV) in blood samples is being extensively studied, as it has the potential to provide clinically meaningful biomarkers for health and disease. This study investigated the impact of different blood collection tubes and processing intervals on EV characteristics in blood samples. The results showed that the choice of blood collection tube and processing interval significantly affected various metrics related to blood sample quality, ex-vivo generation of EV from blood cells, EV recovery, and molecular signatures associated with EV. These findings provide valuable guidance for selecting optimal conditions for EV analysis and contribute to standardizing EV studies.

Summary: This study proposes a novel system for multiplexed analysis of bladder cancer-derived urine exosomes using magnetic microspheres as barcoded microcarriers. The unique structure of the microcarriers allows for controllable movement and high sensitivity detection of exosomes. The noninvasive nature of the platform, combined with its good sensitivity, specificity, low sample consumption, and ease of use, indicates its great potential for bladder cancer diagnosis in clinical applications.

Summary: In addition to its classical role in innate immunity, intracellular complement components, particularly C3, play essential roles in immune defense, T cell homeostasis, and tumor cell proliferation and metastasis. This study demonstrates that C3 is significantly upregulated in PTX-resistant NSCLC cells and silencing C3 sensitizes resistant cells to PTX therapy. C3b translocates into the nucleus and interacts with the SIN3A complex to repress the expression of GADD45A, resulting in decreased PTX-induced apoptosis. These findings highlight the oncogenic properties of C3 and provide a potential therapeutic strategy to overcome PTX resistance.

Summary: Sufficient evidence has linked many different types of cancers and T2D through shared risk factors; however, the underlying mechanisms are not fully understood. Increased levels of a-hydroxybutyrate (a-HB), a metabolite found in diabetes and cancer, including colorectal cancer (CRC), triggers the translocation of lactate dehydrogenase A (LDHA) to the nucleus. This process leads to an increase in TNF-a production, impaired insulin secretion, and exacerbation of CRC and type 2 diabetes. Metformin can interrupt this process by downregulating LDHA expression, thus highlighting its potential in preventing cancer and diabetes.
