Isolation, functional characterization, and biological properties of MCh-AMP1, a novel antifungal peptide from Matricaria chamomilla L.

The antimicrobial activities of natural products have attracted much attention due to the increasing incidence of pathogens that have become resistant to drugs. Thus, it has been attempted to promisingly manage infectious diseases via a new group of therapeutic agents called antimicrobial peptides. In this study, a novel antifungal peptide, MCh-AMP1, was purified by reverse phase HPLC and sequenced by de novo sequencing and Edman degradation. The antifungal activity, safety, thermal, and pH stability of MCh-AMP1 were determined. This peptide demonstrated an antifungal activity against the tested Candida and Aspergillus species with MIC values in the range of 3.33-6.66 mu M and 6.66-13.32 mu M, respectively. Further, physicochemical properties and molecular modeling of MCh-AMP1 were evaluated. MCh-AMP1 demonstrated 3.65% hemolytic activity at the concentration of 13.32 mu M on human red blood cells and 10% toxicity after 48 hr at the same concentration on HEK293 cell lines. The antifungal activity of MCh-AMP1 against Candida albicans was stable at a temperature range of 30-50 degrees C and at the pH level of 7-11. The present study indicates that MCh-AMP1 may be considered as a new antifungal agent with therapeutic potential against major human pathogenic fungi.

Biochemical responses as early and reliable biomarkers of organophosphate and carbamate pesticides intoxication: A systematic literature review

Summary: Inhibition of cholinesterase (ChE) activity has been used as the main diagnostic method for organophosphate (OP) and carbamate pesticides poisoning. However, ChE activity may also be altered due to exposure to other toxicants and medical conditions. This systematic review identified additional biomarkers, such as beta-glucuronidase, neuropathy target esterase, and hematological indicators, that show high sensitivity and accuracy in the diagnosis of OP poisoning.

The role of serum circulating microbial toxins in severity and cytokine storm of COVID positive patients

Summary: The emergence of Covid-19 is a global problem causing increasing mortality rates and lacking verified treatment. The abnormal unrestricted response called cytokine storm, which is triggered by increased amounts of cytokine production, contributes to the severity of Covid-19 and the development of respiratory failure and pneumonia. The interaction between SARS-CoV-2 and endotoxins plays a role in the excessively elevated release of cytokines and the worsening of Covid-19.

Prevalence and genetic characteristics of fusidic acid resistant Staphylococcus aureus clinical isolates: Emergence of t030 strains carrying fusB in Tehran, Iran

Summary: This descriptive cross-sectional study provides insights into the molecular characteristics and antimicrobial resistance of fusidic acid resistant Staphylococcus aureus strains isolated from hospitalized patients in Iran. The prevalence of fusidic acid resistance in S. aureus isolates was found to be 15.1%, with fusB being the most predominant acquired gene. The study highlights the importance of understanding the local prevalence of FA-resistant MRSA for effective treatment, control, and reduction of these isolates.

Tigecycline-Containing Regimens and Multi Drug-Resistant Acinetobacter baumannii: A Systematic Review and Meta-Analysis

Summary: This systematic review and meta-analysis aimed to explore the safety and efficacy of tigecycline for the treatment of multi drug-resistant Acinetobacter baumannii. The results showed that tigecycline had similar efficacy compared to other antibiotics, but it had a higher mortality rate and lower microbiological eradication rate. Therefore, the use of tigecycline for the treatment of multi drug-resistant A. baumannii is not recommended.

Molecular Characteristics of Staphylococcus aureus Strains to Carry Panton-Valentine Leukocidin Genes Isolated from Hospitalized Patients in Tehran, Iran

Summary: This study investigated the genetic characteristics of PVL-positive S. aureus strains isolated from clinical samples. The emergence of vancomycin-resistant S. aureus strains among PVL-positive S. aureus strains in Iran is a serious alarm and poses the greatest concern in the treatment of staphylococcal infections in the healthcare setting. Direct transfers between community and nosocomial PVL-positive S. aureus types were observed.

Genetic diversity of Staphylococcus aureus isolated from ear infections in Iran: Emergence of CC8/ST239-SCCmec III as major genotype

Summary: The study investigated the antibiotic resistance profile and genetic variability of Staphylococcus aureus isolates from ear infections in adult patients in Tehran, Iran. The results showed high resistance to antibiotics and strong biofilm formation ability in these strains. Different genetic differences were observed in strains from different types of ear infections.

Roles of Different β-Defensins in the Human Reproductive System: A Review Study

Summary: Human beta-defensins (hBDs) are cationic peptides with amphipathic spatial shape and high cysteine content. The members of this peptide family have various functions in the human body, including the reproductive system. Beta-defensin 1 interacts with chemokine receptor 6 (CCR6) to prevent bacterial infections in the male reproductive system and has positive effects on antitumor immunity and fertilization in the female reproductive system. Beta-defensin 2 has antibacterial action and may play a role in preventing cervical cancer through interactions with dendritic cells. Beta-defensin 126 is essential for sperm motility and protection against immune system factors. This study reviews the roles of beta-defensin 1, beta-defensin 2, and beta-defensin 126 in both the male and female reproductive systems.

Investigating the toxicity of germ of date seed on normal and cancerous cell line and P53 gene expression

Summary: The study collected the germ of Date seeds by breaking a thousand date kernels, and it was powdered and dissolved in distilled water to achieve different concentrations. Cancerous and normal cell lines were treated with these concentrations for different time periods, and cell viability and P53 gene expression were evaluated. Results showed that the germ of Date seed led to a decrease in viable cells and an increase in P53 gene expression in both cancerous and normal cell lines.

Behavioral studies of mice with breast cancer after treatment with new anticancer agent, Rh2-containing arginine-graphene

Summary: The study evaluated the efficacy of Ginsenoside Rh2 and its combination with arginine-reduced graphene (Gr-Arg-Rh2) in treating mice with breast cancer. The results showed that mice treated with Gr-Arg and Gr-Arg-Rh2 exhibited more energy compared to Rh2 and control groups. Based on these findings, Gr-Arg-Rh2 may reduce cancer-related fatigue and low energy in cancer patients.

Relationship of severity of COVID-19 disease with lung-CT-scan imaging

Summary: This study evaluated lung CT images in COVID-19 patients and analyzed their relationship with clinical and laboratory factors as well as the worst disease condition. The results showed a high prevalence of lung and cardiac involvement. Age was significantly related to recovery. CT score, bilateral lung involvement, and Crazy paving had a significant effect on recovery rate.

Comparison of Rectal and Sublingual Misoprostol Prescription to Reduce Bleeding During and After Cesarean Section: double blind, randomized clinical trial

Summary: The aim of this study was to compare the effectiveness of rectal and sublingual misoprostol administration in reducing bleeding during and after cesarean section. The results showed no significant difference between the two groups in intraoperative and postoperative bleeding, as well as side effects. Therefore, sublingual misoprostol can be used as an alternative to rectal misoprostol in preventing postpartum hemorrhage.

Dihydropyrimidine derivatives as MDM2 inhibitors

Summary: One potential approach for tumor therapy is inhibiting the binding between MDM2 and p53 to reactivate p53 in tumor cells. In this study, Monastrol derivatives were designed as MDM2 inhibitors and evaluated for their cytotoxicity on cancer cells. Compound 5d showed the best inhibitory results in silico and in vitro experiments. These findings suggest that Monastrol derivatives have the potential to be candidates for MDM2 inhibition.
