By using machine learning, we have developed a predictive model for TOP2B binding based on accessible chromatin and architectural factors. This model accurately reflects experimental data, demonstrating the importance of TOP2B for chromatin accessibility and organization.

Summary: RNA editing is a process that alters RNA sequence, with nonsynonymous editing significantly enriched in ubiquitination sites. Previous research found that lysine codons are enriched in the preferred deamination sites for RNA editing. Our study found that mRNA with edited ubiquitination sites can be translated into protein, and these editing sites showed differential editing in tumor samples and were correlated with clinical outcomes. Additionally, these genes were specifically enriched in the immune response pathway.

Summary: This study identified vulnerabilities in Burkitt lymphoma (BL) through genome-scale CRISPR-Cas9 screens and validated serine hydroxymethyltransferase 2 (SHMT2) as a potential drug target. Mechanistically, SHMT2 inhibition disrupted the oncogenic transcription factor TCF3 and collapsed tonic BCR signaling, providing new perspectives for innovative therapies.
