African swine fever: an unprecedented disaster and challenge to China

BackgroundAfrican swine fever (ASF), caused by African swine fever virus, is a hemorrhagic and often fatal disease of domestic pigs and wild boar, which is notifiable to the World Organization for Animal Health. On August 3, 2018, China reported the first outbreak of ASF in Shenyang, a northeastern city of China. As of October 8, a total of 33 ASF outbreaks were reported in eight provinces in China, the biggest pork producer and consumer in the world.Main bodyThis commentary summarizes the current situation of ASF in China, measures that China has taken to control the disease, lessons learnt from other countries, challenges and recommendations on ASF control in China, and possible international collaborations on ASF.ConclusionsASF is an unprecedented disaster and challenge to the Chinese swine industry. It will be a formidable and protracted campaign to control ASF in China, which requires joint participation and coordination of stakeholders and agencies at different levels.

Adaptation of African swine fever virus to HEK293T cells

Summary: The study revealed that African swine fever virus undergoes genomic variations during adaptation to different cell lines, with deletions in MGF300 and MGF360 genes playing a crucial role in early adaptation to HEK293T cells. However, after adaptation, the virus displayed reduced infectivity in primary porcine alveolar macrophages compared to the wild-type strain. Further research is needed to understand the biological functions of the deletions and mutations associated with ASFV infection in different cells and animals.

Current efforts towards safe and effective live attenuated vaccines against African swine fever: challenges and prospects

Summary: The major risks of ASF spread are insufficient biosecurity measures and human behaviors. Therefore, a safe and effective vaccine seems to be a reasonable demand for the prevention and control of ASF. Several critical issues need to be addressed to commercialize an ideal ASF LAV, including a stable cell line for manufacturing vaccines, differentiation of infected from vaccinated animals (DIVA), and cross-protection from different genotypes.

The A137R Protein of African Swine Fever Virus Inhibits Type I Interferon Production via the Autophagy-Mediated Lysosomal Degradation of TBK1

Summary: African swine fever (ASF) is a lethal viral disease of pigs caused by African swine fever virus (ASFV). A recent study found that deleting the A137R gene in ASFV attenuates its virulence and promotes the expression of type I interferon (IFN) in primary porcine alveolar macrophages. The A137R protein negatively regulates the cGAS-STING-mediated IFN-beta signaling pathway through the autophagy-mediated lysosomal degradation of TBK1. This study enhances the understanding of ASFV immunoevasion strategies and provides new insights for the development of live attenuated ASF vaccines.

Construction of Attenuated Strains for Red-Spotted Grouper Nervous Necrosis Virus (RGNNV) via Reverse Genetic System

Summary: In this study, a reverse genetic system for the culture of recombinant red-spotted grouper nervous necrosis virus (RGNNV) is developed for the first time. Attenuated RGNNV strains with the loss of B2 protein expression were constructed and compared in terms of growth rate, Mx1 expression, and virulence. This study provides a potential tool for further research on viral protein function, virulence pathogenesis, and vaccine development of RGNNV.

The 24.5-kb Left Variable Region Is Not a Determinant for African Swine Fever Virus to Replicate in Primary Porcine Alveolar Macrophages

Summary: This study demonstrated that the ASFV-P121 strain, which has a deletion in the left variable region (LVR), lost the ability to replicate in primary porcine alveolar macrophages (PAMs). However, a mutant virus with the same LVR deletion only showed a 10-fold reduction in growth compared to the wild-type virus. Importantly, this large deletion did not affect virus factory formation or virion morphogenesis. These findings provide insights into the molecular mechanism of ASFV cell tropism change.

Summary: In this study, MGF300-2R was identified as a key factor for replication and pathogenicity of African swine fever virus (ASFV). MGF300-2R promotes the K27-linked ubiquitination of IKKα and IKKβ for TOLLIP-dependent autophagic degradation, suppressing the activation of the NF-κB signaling pathway. These findings highlight the relevance of the inflammatory response in ASFV virulence and suggest MGF300-2R as a potential target for vaccine development.

Summary: This study reveals the essential role of the ASFV structural protein pH240R in virus morphogenesis. pH240R interacts with the major capsid protein p72 and is critical for the assembly of infectious particles. Deletion of pH240R leads to reduced progeny virus titer and increased production of noninfectious particles. Furthermore, pH240R deficiency induces high-level expression of inflammatory cytokines. These findings provide insights into ASFV assembly and offer a potential target for the development of vaccines and antivirals.
