Synergistic Activity of Niclosamide in Combination With Colistin Against Colistin-Susceptible and Colistin-Resistant Acinetobacter baumannii and Klebsiella pneumoniae

Colistin is among the few antibiotics effective against multidrug-resistant Acinetobacter baumannii and Klebsiella pneumoniae clinical isolates. However, in the last few years, colistin-resistant A. baumannii and K. pneumoniae strains have emerged. Therefore, combination therapies, between colistin and other old drugs, restoring the activity of colistin are required. The main objective of this study was to analyse the activity of niclosamide, an anthelmintic drug, in combination with colistin against colistin-susceptible (Col-S) and colistin-resistant (Col-R) A. baumannii and K. pneumoniae. The MIC were determined by microdilution assay and the time-kill curves were performed. The zeta potential of Col-S and Col-R of A. baumannii and K. pneumoniae in presence of niclosamide was assessed. Niclosamide in combination with colistin showed improved activity against Col-S and Col-R A. baumannii and K. pneumoniae. Time-killing curves showed synergic activity between niclosamide and colistin against Col-S and Col-R A. baumannii and K. pneumoniae, especially when niclosamide or colistin was added for second time at 4 h of the 24 h killing curve. Col-R A. baumannii and K. pneumoniae in presence of niclosamide exhibited a greater negative charge (-34.95 +/- 0.35 mV and -38.85 +/- 0.92 mV; P < 0.05) than Col-R A. baumannii and K. pneumoniae in absence of niclosamide (-26.85 +/- 3.65 mV and -35.27 +/- 0.72 mV). These data suggest that niclosamide might be combined with colistin, being a potential alternative for treatment of Col-R Gram-negative bacilli infections.

Analysis of bacterial pangenomes reduces CRISPR dark matter and reveals strong association between membranome and CRISPR-Cas systems

Summary: CRISPR-Cas systems are a type of acquired immunity found in 40% of bacterial genomes, which use small DNA fragments called spacers to prevent viral infections. However, the majority of these spacers, known as CRISPR dark matter, have not been associated with specific viruses. Through analyzing spacers from tens of thousands of genomes, researchers have reduced the amount of CRISPR dark matter from 80% to as low as 15% in some bacterial species. Additionally, it has been observed that genomes with CRISPR-Cas systems also have specific sets of membrane proteins, suggesting a connection between bacterial competitiveness and the acquisition of CRISPR-Cas.

Role of bla(TEM) and OmpC in the piperacillin-tazobactam resistance evolution by E. coli in patients with complicated intra-abdominal infection

Summary: This study aimed to identify the mechanisms underlying Piperacillin-tazobactam resistance (P/T-R) in Escherichia coli. The results showed that P/T-R isolates had a higher copy number of the bla(TEM) gene, generated by three different genetic events. In addition, reduced expression of OmpC was observed in two P/T-R strains. This study provides important insights into the acquisition of P/T-R in E. coli, which is crucial for effective treatment and prevention of resistant microorganisms.

Summary: West Nile virus (WNV) is a neurotropic flavivirus transmitted by mosquitoes and can cause severe diseases. By studying experimentally infected mice and naturally infected patients, dynamic alterations in the lipidome were identified as metabolic fingerprints of different infection stages. The dysregulation of sphingolipid metabolism by WNV may provide new therapeutic opportunities and support the potential of certain lipids as novel peripheral biomarkers of WND progression.
