Summary: Left ventricular non-compaction cardiomyopathy is a rare heart disease characterized by a two-layer structure of the myocardium and increased trabeculae. Studying familial forms of LVNC can help in predicting risks and offering genetic counseling to relatives. In a family with LVNC, two likely pathogenic mutations were identified through genetic testing, and in vitro experiments confirmed the pathogenicity of one of the mutations.

Spatial transcriptomics unveils ZBTB11 as a regulator of cardiomyocyte degeneration in arrhythmogenic cardiomyopathy

Summary: This study used Tomo-Seq technology to create a gene expression atlas of explanted ACM hearts, allowing the identification of new genes and pathways involved in specific remodeling processes. The study found that Zinc finger and BTB domain-containing protein 11 (ZBTB11) is specifically enriched at sites of active fibro-fatty replacement of myocardium, and its overexpression induces autophagy and cell death-related gene programs in human cardiomyocytes. ZBTB11 is identified as a novel driver of cardiomyocyte loss.

Summary: This study reported a patient with muscular dystrophy and early onset dilated cardiomyopathy who carried compound heterozygous variants in FKTN. The research demonstrated that compound heterozygous mutations in FKTN result in loss of fully glycosylated alpha-dystroglycan, leading to cardiomyopathy and end-stage heart failure at a young age.

Altered Expression of TMEM43 Causes Abnormal Cardiac Structure and Function in Zebrafish

Summary: In this study, transgenic zebrafish models were used to investigate the pathogenesis of TMEM43-associated cardiomyopathy. These results provide new insights into the development of TMEM43-associated cardiomyopathy.

Summary: In this study, we found that several variants at specific positions in the prodomain of cardiac desmosomal cadherins can influence intracellular transport. However, some commonly listed variants in genetic databases do not affect intracellular transport. These prodomain variants may be relevant in the context of arrhythmogenic cardiomyopathy.

The Consideration of Pseudoxanthoma Elasticum as a Progeria Syndrome

Summary: By conducting experiments on dermal fibroblasts from healthy individuals and PXE patients, it was found that the gene expression of lamin A and C decreased significantly in PXE fibroblasts, while the gene expression of the aging-related factors progerin and farnesyltransferase increased significantly. These results suggest that the aging processes in PXE patients may be related to other factors rather than defects in the nuclear envelope or nucleoli.

Summary: This study analyzed pediatric myocarditis patients with the dilated cardiomyopathy phenotype using whole-exome sequencing. They found likely pathogenic variants in cardiomyopathy genes and immune disorder genes, suggesting a genetic impact on pediatric myocarditis with the dilated cardiomyopathy phenotype. The presence of heterozygous variants in immune-related genes may also play a role in promoting myocarditis.
