Oleuropein, the Main Polyphenol of Olea europaea Leaf Extract, Has an Anti-Cancer Effect on Human BRAF Melanoma Cells and Potentiates the Cytotoxicity of Current Chemotherapies

Oleuropein (Ole), a secoiridoid glucoside present in Olea europaea leaves, gained scientific interest thanks to its several biological properties, including the anticancer one. We verified whether Ole might potentiate the cytotoxicity of conventional drugs used to treat melanoma, disclosing a potentially new therapeutic strategy. We tested the cytotoxic action of Ole alone or in combination with chemotherapeutics on A375 human melanoma cells. We found that Ole was able, at a dose of 500 mu M, to stimulate apoptosis, while at a non-toxic dose of 250 mu M, it affected cell proliferation and induced the downregulation of the pAKT/pS6 pathway. A dose of 250 mu M Ole did not potentiate the effect of Vemurafenib (PLX4032), but it succeeded in increasing the cytotoxic effect of Dacarbazine (DTIC). The major effect was found in the association between Ole and Everolimus (RAD001), also on PLX4032-resistant BRAF melanoma cells, which possibly cooperate in the inhibition of the pAKT/pS6 pathway. Of interest, an olive leaf extract enriched in equimolar Ole was more effective and able to further improve DTIC and RAD001 efficacy on BRAF melanoma cells with respect to Ole alone. Therefore, Ole represents a natural product able to potentiate a wide array of chemotherapeutics against BRAF melanoma cells affecting the pAKT/pS6 pathway.

A novel conjugate targeting the α v β 6 integrin receptor, linked to the kinase inhibitor nintedanib, inhibits cancer cell adhesion and growth through selective delivery. The conjugate also suppresses clonogenic ability and other aspects associated with EMT transition, offering a precise strategy against non-small cell lung cancer.

European survey of newborn bloodspot screening for CF: opportunity to address challenges and improve performance

Summary: The study aimed to assess the current status and performance of newborn bloodspot screening (NBS) for CF in Europe. A survey was conducted among representatives of NBS programs across Europe, and performance was evaluated using a previously developed framework. The results showed that in 2022, there were 22 national and 34 regional NBS programs in Europe. The integration of DNA analysis into the NBS protocol improved the positive predictive value, but also led to increased carrier and CFSPID recognition.

Oxidative Stress and Natural Antioxidants in Osteoporosis: Novel Preventive and Therapeutic Approaches

Summary: This review provides a detailed report on the cellular and molecular mechanisms involved in bone remodeling, specifically in relation to oxidative stress, inflammatory factors, and estrogen deficiency. Oxidative stress is a key factor in the development of osteoporosis, leading to osteocyte apoptosis and impaired bone remodeling and resorption. Recent studies have shown that natural antioxidants found in the diet can effectively prevent and reduce the negative effects of oxidative stress on bone health. These antioxidants target osteocytes and their molecular factors, promoting bone regeneration and preventing bone loss. This suggests that incorporating antioxidants into therapeutic approaches may improve the prevention and treatment of osteoporosis and related bone diseases.

Summary: Newborn screening for inborn errors of metabolism is an advanced tool for early diagnosis and treatment initiation. Italy introduced expanded newborn screening in 2016-2017 and collected data from 15 Italian screening laboratories. Aminoacidemias were the most common inborn errors, and second-tier tests were effective in reducing false positives. The screening also identified conditions related to maternal deficiencies.

However, the availability and conduct of NBS programmes vary across Europe, highlighting the need for improvement to ensure equity and high-quality screening.
