Inhibition of endoplasmic reticulum stress protected DOCA-salt hypertension-induced vascular dysfunction

Hypertension has complex vascular pathogenesis and therefore the molecular etiology remains poorly elucidated. Endoplasmic reticulum stress (ERS), which is a condition of the unfolded/misfolded protein accumulation in the endoplasmic reticulum, has been defined as a potential target for cardiovascular disease. In the present study, the effects of ERS inhibition on hypertension-induced alterations in the vessels were investigated. In male Wistar albino rats, hypertension was induced through unilateral nephrectomy, deoxycorticosteroneacetate (DOCA) injection (20 mg/kg, twice a week) and 1% NaCl with 0.2% KCI added to drinking water for 12 weeks. An ERS inhibitor, tauroursodeoxycolic acid (TUDCA) (150 mg/kg/day, i.p.), was administered for the final four weeks. ERS inhibition in DOCA-salt induced hypertension was observed to have reduced systolic blood pressure, improved endothelial dysfunction, enhanced plasma nitric oxide (NO) level, reduced protein expressions of phosphorylated-double-stranded RNA-activated protein kinase-like endoplasmic reticulum kinase (pPERK), 78 kDa glucose-regulated protein (GRP78), Inositol trisphosphate receptor1 (IP(3)R1) and Epidermal growth factor receptor (EGFR), increased expressions of endoplasmic reticulum Ca2+-ATPase2 (SERCA2) and B cell lymphoma2 (Bcl2) in vessels. These findings suggest that the beneficial effects of ERS inhibition on hypertension may be related to protection of vessel functions through restoration of endoplasmic reticulum calcium homeostasis, and apoptotic and mitotic pathways.

Lactobacillus plantarum improves lipogenesis and IRS-1/AKT/eNOS signalling pathway in the liver of high-fructose-fed rats

Summary: The present study investigated the effects of Lactobacillus plantarum and Lactobacillus helveticus supplementation on liver lipogenesis, insulin signaling, and glucose transporters in high-fructose-fed rats. The results showed that supplementation with Lactobacillus plantarum improved liver health by reducing weight, lipid accumulation, and FASN expression, while also improving IRS-1/AKT/eNOS pathway and GLUT2 expression. On the other hand, Lactobacillus helveticus supplementation had a restoring effect on lipid accumulation by decreasing FASN expression and regulating IRS-1 and GLUT2 expressions.

Summary: This study aimed to evaluate the impact of high-fructose corn syrup (HFCS) intake on antioxidative structures and inflammation in the gingival tissue and masseter muscle of rats. The results showed that HFCS increased proinflammatory structures and suppressed antioxidant enzymes in these tissues, while kefir supplementation improved these undesirable changes. This study suggests that kefir consumption may have restorative effects on oral health disorders caused by high levels of fructose in the diet.

Summary: This study compared the effects of resveratrol supplementation and regular exercise on hypertension-induced cellular stress responses of the myocardium. The results showed that both resveratrol and exercise reduced systolic blood pressure in hypertensive rats, improved cardiac muscle responses, and reduced oxidative stress. Regular exercise restored endoplasmic reticulum stress and decreased mitogenic activation, while resveratrol only improved mitophagy. Additionally, both resveratrol and exercise suppressed inflammation.

Aging and gender-related effects of tauroursodeoxycholic acid treatment on liver functions, plasma lipid profile, and oxidative stress

Summary: This study reveals the effects of the ER stress inhibitor TUDCA on liver function, plasma lipids, and oxidative stress markers in the elderly. The results showed that TUDCA decreased AST levels, increased HDL value, decreased TG value, and reduced MDA and MPO levels in the elderly. The effects on some parameters varied depending on gender.

The impact of dietary fructose on gut permeability, microbiota, abdominal adiposity, insulin signaling and reproductive function

Summary: Excessive intake of fructose in the regular human diet is associated with global increases in metabolic disorders. Sugar-sweetened soft drinks, the main source of added fructose, can lead to gut permeability, inflammatory response, hepatic steatosis, abdominal adiposity, insulin resistance, and negative impacts on reproductive function. Recognizing the detrimental effects of fructose and implementing relevant public health policies are essential for preventing diet-related metabolic disorders.

Evaluation of Biological Activities and Enzyme Inhibitory Potential of 7,8-Dihydroxy-3-(4-methylphenyl) Coumarin

Summary: This study investigated the antioxidant, antibacterial, cytotoxic, and antioxidant enzyme inhibitory properties of 7,8-dihydroxy-3-(4-methylphenyl) coumarin. It exhibited high free radical scavenging activity, total antioxidant capacity, and reducing power. The compound also demonstrated antibacterial effects and cytotoxic activity on breast cancer cells, while being non-toxic to liver cancer cells. Furthermore, it inhibited catalase enzyme and glutathione-S-transferase activities. These findings suggest the compound's potential in preventing oncogenesis and tumor progression in the liver and breast cancer.
