Naringenin Attenuates Myocardial Ischemia-Reperfusion Injury via cGMP-PKGI alpha Signaling and In Vivo and In Vitro Studies

Endoplasmic reticulum (ER) stress and oxidative stress contribute greatly to myocardial ischemia-reperfusion (MI/R) injury. Naringenin, a flavonoid derived from the citrus genus, exerts cardioprotective effects. However, the effects of naringenin on ER stress as well as oxidative stress under MI/R condition and the detailed mechanisms remain poorly defined. This study investigated the protective effect of naringenin on MI/R-injured heart with a focus on cyclic guanosine monophosphate- (cGMP-) dependent protein kinase (PKG) signaling. Sprague-Dawley rats were treated with naringenin (50 mg/kg/d) and subjected to MI/R surgery with or without KT5823 (2 mg/kg, a selective inhibitor of PKG) cotreatment. Cellular experiment was conducted on H9c2 cardiomyoblasts subjected to simulated ischemia-reperfusion treatment. Before the treatment, the cells were incubated with naringenin (80 mu mol/L). PKGI alpha siRNA was employed to inhibit PKG signaling. Our in vivo and in vitro data showed that naringenin effectively improved heart function while it attenuated myocardial apoptosis and infarction. Furthermore, pretreatment with naringenin suppressed MI/R-induced oxidative stress as well as ER stress as evidenced by decreased superoxide generation, myocardial MDA level, gp91(phox) expression, and phosphorylation of PERK, IRE1 alpha, and EIF2 alpha as well as reduced ATF6 and CHOP. Importantly, naringenin significantly activated myocardial cGMP-PKGI alpha signaling while inhibition of PKG signaling with KT5823 (in vivo) or siRNA (in vitro) not only abolished these actions but also blunted naringenin's inhibitory effects against oxidative stress and ER stress. In summary, our study demonstrates that naringenin treatment protects against AIM injury by reducing oxidative stress and ER stress via cGMP-PKGI alpha signaling. Its cardioprotective effect deserves further clinical study.

Protective effects and mechanisms of psoralidin against adriamycin- induced cardiotoxicity

Summary: This study provides evidence for the beneficial effect of psoralidin (PSO) on adriamycin (ADR)-induced cardiotoxicity through the activation of the SIRT1/PPARy signaling pathway. PSO improves cardiac function and mitochondrial function, reduces myocardial fibrosis, oxidative stress, and apoptosis. These findings suggest that PSO could be a potential cardioprotective drug candidate for mitigating ADR-induced cardiotoxicity.

Assessing left atrial function in patients with atrial fibrillation and valvular heart disease using cardiovascular magnetic resonance imaging

Summary: This study evaluated the left atrial function in patients with atrial fibrillation associated with valvular heart disease using cardiovascular magnetic resonance imaging feature tracking. The results showed decreased left atrial strain parameters and increased left atrial size and volume in the patient group compared to healthy controls. The study also found a significant correlation between decreased left atrial total ejection fraction and altered strain and strain rate. The findings suggest that cardiovascular magnetic resonance imaging feature tracking can be a helpful noninvasive tool for assessing left atrial function in the clinical treatment of atrial fibrillation associated with valvular heart disease.

Preventive Effect of Berberine on Postoperative Atrial Fibrillation

Macrophage Rmp Ameliorates Myocardial Infarction by Modulating Macrophage Polarization in Mice

Summary: This study investigated the role of macrophage RNA polymerase II subunit 5-mediating protein (RMP) in myocardial infarction (MI). The results showed that RMP deficiency worsened cardiac dysfunction and promoted M1 polarization while inhibiting angiogenesis after MI. In contrast, RMP overexpression in macrophages promoted M2 polarization and angiogenesis after MI. Mechanistically, RMP regulated macrophage polarization through the HSP90-p38 signaling pathway.

Protection of melatonin treatment and combination with traditional antibiotics against septic myocardial injury

Summary: This study investigates the effects and mechanisms of melatonin pretreatment, posttreatment, and combination with antibiotics in the treatment of sepsis and septic myocardial injury. Results show that melatonin pretreatment has a protective effect on sepsis and septic myocardial injury, related to attenuation of inflammation and oxidative stress, improvement of mitochondrial function, regulation of endoplasmic reticulum stress, and activation of the AMPK signaling pathway. Melatonin posttreatment also has a certain degree of protection, while the combination with antibiotics has limited effect. This study provides a theoretical basis for the application strategy and combination of melatonin in septic myocardial injury.
