Total Synthesis of Emmyguyacins A and B, Potential Fusion Inhibitors of Influenza Virus

Fungal glycolipids emmyguyacins A and B inhibit the pH-dependent conformational change of hemaglutinin A during replication of the Influenza virus. Herein, we report the first total synthesis and structure confirmation of emmyguyacins A and B. Our efficient route, which involves regioselective functionalization of trehalose, allows rapid access to adequate amounts of chemically pure emmyguyacin analogues including the desoxylate derivatives for SAR studies.

Unusual C-C bond cleavage of an α-trifloxy Sialic acid hemiacetal under Lattrell-Dax conditions

Summary: This study describes a novel oxidative fragmentation reaction of methyl ester in DMF with sodium nitrite, yielding a new compound with high yield. Furthermore, another new compound is obtained through the reaction of the same ester with sodium carbonate. Reduction and acetylation of the lactone result in a peracetylated aminodeoxyheptitol with a zig zag conformation of its carbon backbone.

Synthesis of Gentamicin Minor Components: Gentamicin C1a and Gentamicin C2b

Summary: In this study, we report a straightforward synthesis of Gentamicin C1a and the minor isomer C2b from readily available sisomicin. These compounds exhibit favorable properties in terms of antibacterial activity and toxicity compared to the commercial mixture.

Total Synthesis of a Trehalose-Containing Lipooligosaccharide Analogue from Mycobacterium linda

Summary: This article introduces a simple method for synthesizing a lipooligosaccharide analogue from Mycobacterium linda isolated from Crohn's disease. The total synthesis of the tetrasaccharide was achieved through a [2+2] glycosylation reaction, with the key features being the selective functionalization of a trehalose core via highly regioselective acylations and regioselective glycosylations. The synthesis was completed in a longest linear sequence of 14 steps with an overall yield of 14.2%.

Synthesis of Gentamicins C1, C2, and C2a and Antiribosomal and Antibacterial Activity of Gentamicins B1, C1, C1a, C2, C2a, C2b, and X2

Summary: In this study, we synthesized gentamicins C1, C2, and C2a with methyl substitution at the 6'-position, providing an alternative method for accessing these compounds compared to previous chromatographic methods. We also explored the antiribosomal activity of the synthetic gentamicin congeners against bacterial ribosomes and different types of ribosomes, establishing structure-activity relationships with the substitution pattern around ring I, antibacterial resistance, and cochlear toxicity data from literature.
