BMI1 is a therapeutic target in recurrent medulloblastoma

Medulloblastoma (MB) is the most frequent malignant pediatric brain tumor, representing 20% of newly diagnosed childhood central nervous system malignancies. Although advances in multimodal therapy yielded a 5-year survivorship of 80%, MB still accounts for the leading cause of childhood cancer mortality. In this work, we describe the epigenetic regulator BMI1 as a novel therapeutic target for the treatment of recurrent human Group 3 MB, a childhood brain tumor for which there is virtually no treatment option beyond palliation. Current clinical trials for recurrent MB patients based on genomic profiles of primary, treatment-naive tumors will provide limited clinical benefit since recurrent metastatic MBs are highly genetically divergent from their primary tumor. Using a small molecule inhibitor against BMI1, PTC-028, we were able to demonstrate complete ablation of self-renewal of MB stem cells in vitro. When administered to mice xenografted with patient tumors, we observed significant reduction in tumor burden in both local and metastatic compartments and subsequent increased survival, without neurotoxicity. Strikingly, serial in vivo re-transplantation assays demonstrated a marked reduction in tumor initiation ability of recurrent MB cells upon re-transplantation of PTC-028-treated cells into secondary recipient mouse brains. As Group 3 MB is often metastatic and uniformly fatal at recurrence, with no current or planned trials of targeted therapy, an efficacious targeted agent would be rapidly transitioned to clinical trials.

Comparison of Two Drainage Systems on Chronic Subdural Hematoma Recurrence

Summary: This study retrospectively analyzed data from 172 patients with chronic subdural hematoma (CSDH) and compared the effect of two drainage systems on the recurrence rate of CSDH. The results showed no significant difference in the recurrence rate between the use of a pediatric size nasogastric tube and an external ventricular drainage catheter.

Summary: The purpose of this study was to determine whether intraoperative adjunctive EVD placement in patients with a posterior fossa tumor (PFT) led to improved surgical, radiographic, and clinical outcomes. The results showed that the routine use of EVD as an intraoperative adjunct in clinically stable pediatric patients with posterior fossa tumors and hydrocephalus may not be associated with improved radiological or clinical outcomes.

A single center experience in the management of progressive juvenile pilocytic astrocytoma

Summary: Brain metastases (BM) are the most common brain neoplasm in adults, but current therapies have limited efficacy. This study analyzed the transcriptional profile of brain metastasis initiating cells (BMICs) at two distinct stages of the metastatic cascade. The researchers found that premetastatic BMICs exhibited increased expression of HLA-G, which plays a role in promoting brain metastatic lesions. Understanding the molecular landscape of premetastatic BMICs could lead to the development of more effective therapies.

Current treatment mainly involves surgical interventions, but they have complications and are not ideal for preserving fertility. This study developed biocompatible albendazole-loaded solid lipid nanoparticles with sustained-release properties, which enhanced the bioavailability of the drug and showed potential as an effective anthelmintic drug delivery vehicle.

Chain Ratio Type Exponential Estimator for Finite Population Mean in Double Sampling

Summary: This paper proposes a new chain ratio type exponential estimator for finite population mean in double sampling, extending the Chand (1975) chain ratio estimator using exponential strategy. The bias and mean squared error of the developed estimator are derived up to the first degree of approximation. The properties of the estimator are examined and compared with other estimators through theoretical and empirical analysis using simulation study.

Summary: 4H-Chromene derivatives are potential anticancer compounds that bind to the colchicine binding site of tubulin protein. The 2-amino-4-phenyl-4H-benzo[h]chromene-3-carbonitrile-based 4H-chromenes scaffold has a significant impact on tubulin proteins. Understanding the interaction pattern of these compounds against various tubulin isoforms is important for selective targeting of cancer cells. Structural modification can potentially enhance the selective targeting ability of these molecules.

Intracellular Zn(ii) induced turn-on fluorescence of an l-phenylalanine-derived pseudopeptide

Summary: A C-2 symmetric l-phenylalanine-derived pseudopeptide has been synthesized for selective and sensitive recognition of Zn(ii) ions in aqueous-organic media. The pseudopeptidic probes exhibit intracellular Zn(ii) ion-sensing capabilities as demonstrated via live-cell fluorescence studies on RAW264.7 cells. Therefore, this study presents a bioinspired pseudopeptide for potential biological applications involving intracellular Zn(ii) ion detection.
