Interplay between small and long non-coding RNAs in cutaneous melanoma: a complex jigsaw puzzle with missing pieces

The incidence of cutaneous melanoma (CM) has increased in the past few decades. The biology of melanoma is characterized by a complex interaction between genetic, environmental and phenotypic factors. A greater understanding of the molecular mechanisms that promote melanoma cell growth and dissemination is crucial to improve diagnosis, prognostication, and treatment of CM. Both small and long non-coding RNAs (lncRNAs) have been identified to play a role in melanoma biology; microRNA and lncRNA expression is altered in transformed melanocytes and this in turn has functional effects on cell proliferation, apoptosis, invasion, metastasis, and immune response. Moreover, specific dysregulated ncRNAs were shown to have a diagnostic or prognostic role in melanoma and to drive the establishment of drug resistance. Here, we review the current literature on small and lncRNAs with a role in melanoma, with the aim of putting into some order this complex jigsaw puzzle.

Summary: This study describes the clinical characteristics of a cohort of Italian patients with aBCC and investigates the effectiveness and safety of HHI. The results demonstrate that HHI is effective and safe in this patient population and replicate the results of pivotal trials in a real-life clinical setting.

Summary: Immunotherapy in oncology is replacing traditional therapies due to its specific action and limited side effects. However, bacterial infections can still occur as side effects. This case report highlights the importance of considering lifestyle and cutaneous background before starting immunotherapy for cancer treatment, with a focus on pharmacogenomics and the possibility of modified skin microbiota predisposing to cutaneous infections in patients treated with PD-1 inhibitors.

Follicular colonization in melanocytic nevi and melanoma: A literature review

Summary: This article discusses the follicular colonization in melanoma and melanocytic nevi, as well as the related facts and controversies. The lentiginous spread of melanocytes into the hair follicle is observed in various benign melanocytic neoplasms such as nevi, sun-induced melanocytic hyperplasia, and melanoma. The role of follicular colonization in melanoma pathologic staging is still debated, despite its description being recommended by the International Collaboration on Cancer Reporting.

Summary: This study reported a case series of cutaneous melanoma (CM) with p16 overexpression and analyzed their clinicopathologic features, immunohistochemical expression of cell cycle regulatory molecules, and mutational landscape. The results showed that p16 overexpression is a rare event occurring in advanced-stage, clinically and histologically heterogeneous CM. The cases could be classified into three groups based on the expression of cell cycle regulatory molecules and mutational profiles, and zonal p16 overexpression may indicate molecular progression.

PPM1D activity promotes the replication stress caused by cyclin E1 overexpression

Summary: Oncogene-induced replication stress is a major cause of genome instability in cancer cells. This study reveals that increased activity of PPM1D exacerbates replication stress caused by cyclin E1 overexpression, leading to abnormal cell cycle progression and accumulation of DNA copy number alterations. Pharmacological inhibition of PPM1D can alleviate replication stress-induced genome instability.

Summary: High-grade serous ovarian cancer (HGSOC), the deadliest gynecological malignancy, spreads through transcoelomic dissemination. This study reveals that platelet-derived growth factor receptor beta (PDGFRβ) is essential for the formation of tumorspheres in HGSOC. Inhibition of PDGFRβ blocks the clustering of ovarian cancer cells and prevents peritoneal dissemination.
