Identification of the Ferric-Acinetobactin Outer Membrane Receptor in Aeromonas salmonicida subsp salmonicida and Structure-Activity Relationships of Synthetic Acinetobactin Analogues

Aeromonas salmonicida subsp. salmonicida, the causative agent of furunculosis in several fish species, produces acinetobactin and amonabactin as siderophores. In a previous study, we chemically characterized these siderophores and proposed a biosynthetic pathway based on genetic analysis. However, the internalization mechanisms of ferric-acinetobactin and ferric-amonabactin remain largely unknown. In the present study, we demonstrate that the outer membrane protein FstB is the ferric-acinetobactin receptor in A. salmonicida since an fstB defective mutant is unable to grow under iron limitation and does not use acinetobactin as an iron source. In order to study the effect that structural changes in acinetobactin have on its siderophore activity, a collection of acinetobactin-based analogues was synthesized, including its enantiomer and four demethylated derivatives. The biological activity of these analogues on an fstB(+) strain compared to an fstB(-) strain allowed structure-activity relationships to be elucidated. We found a lack of enantiomer preference on the siderophore activity of acinetobactin over A. salmonicida or on the molecular recognition by FstB protein receptor. In addition, it was observed that A. salmonicida could not use acinetobactin analogues when imidazole or a similar heterocyclic ring was absent from the structure. Surprisingly, removal of the methyl group at the isoxazolidinone ring induced a higher biological activity, thus suggesting alternative route(s) of entry into the cell that must be further investigated. It is proposed that some of the synthetic acinetobactin analogues described here could be used as starting points in the development of novel drugs against A. salmonicida and probably against other acinetobactin producers like the human pathogen Acinetobacter baumannii.

Antifungal Potential of Marine Organisms of the Yucatan Peninsula (Mexico) against Medically Important Candida spp.

Summary: This study screened 65 extracts from marine organisms of the Yucatan Peninsula, Mexico, for antifungal activity and identified nine crude extracts with potent antifungal activity, including four from sponge species. The chemical composition analysis of one of the most active fractions revealed the presence of mirabilin B and penaresidin B. These findings highlight the marine organisms of the Yucatan Peninsula as important sources of natural products with promising fungicidal activity, which could greatly advance the treatment of invasive fungal infections, particularly in immunosuppressed patients.

Summary: This study successfully performed stereo-selective total synthesis of Pcb analogues, which are siderophores produced by pathogenic Gram-negative bacteria. The replacement of the acid-sensitive α-methylthiazoline moiety with a more stable thiazole ring and varying the OH group configuration at C-13 resulted in different metal coordination abilities. The study also confirmed the stereochemistry of the Ga3+ complex and proposed the scaffold's potential use in designing new chelating agents or antibacterials.

Summary: Natural Products (NP) play a crucial role in the discovery of novel drugs and biotechnological applications. However, the process of NP discovery is expensive and time-consuming, mainly due to the challenges of dereplication and structure elucidation, especially for metabolites with stereogenic centers. This review highlights recent technological and instrumental advances that address these obstacles and accelerate NP discovery for biotechnological applications. It emphasizes innovative high-throughput tools and methods for bioactivity screening, NP chemical analysis, dereplication, metabolite profiling, metabolomics, genome sequencing and/or genomics approaches, databases, bioinformatics, chemoinformatics, and three-dimensional NP structure elucidation.

Low salinity activates a virulence program in the generalist marine pathogen Photobacterium damselae subsp. damselae

Summary: This study investigated the transcriptional response of the facultative marine pathogen Photobacterium damselae subsp. damselae to changes in salinity. It was found that low salinity conditions triggered a virulence gene expression profile, while growth at high salinity upregulated genes involved in energy production and metabolism. This study enhances our understanding of the salinity-responsive adaptations of a generalist marine pathogen.

Multistage Detection of Tetrodotoxin Traces in Diodon hystrix Collected in El Salvador

Summary: This study presents a multistage methodology for detecting small amounts of tetrodotoxin in fish and potentially other marine organisms. The methodology was applied to porcupinefish specimens collected in El Salvador. It involved a three-stage approach, including sample screening, high-resolution mass spectrometry analyses, and further analysis using different techniques. This study reports the presence of tetrodotoxin in D. hystrix collected in El Salvador for the first time.

Synergistic antileishmanial activity of erythrodiol, uvaol, and oleanolic acid isolated from olive leaves of cv. Chemlali

Summary: This study assessed the antileishmanial activity of biomolecules obtained from Olea europaea L. leaves and twigs from different Tunisian cultivars. The cyclohexane fraction of cv. Chemlali showed the strongest leishmanicidal activity against all tested leishmanial strains. Three triterpenic compounds were identified, including oleanolic acid, erythrodiol, and uvaol, which demonstrated antileishmanial activity. The mixture of these compounds exhibited enhanced activity against Leishmania tropica compared to individual compounds.
