Background Masseter muscle function influences mandibular bone homeostasis. As previously reported, bone resorption markers increased in the mouse mandibular condyle two days after masseter paralysis induced with botulinum toxin type A (BoNTA), followed by local bone loss. Objective This study aimed to evaluate the bone quality of both the mandibular condyle and alveolar process in the mandible of adult mice during the early stage of a BoNTA-induced masseter muscle atrophy, using a combined 3D histomorphometrics and shape analysis approach. Methods Adult BALB/c mice were divided into an untreated control group and an experimental group; the latter received one single BoNTA injection in the right masseter (BoNTA-right) and saline in the left masseter (Saline-left). 3D bone microstructural changes in the mandibular condyle and alveolar process were determined with high-resolution microtomography. Additionally, landmark-based geometric morphometrics was implemented to assess external shape changes. Results After 2 weeks, masseter mass was significantly reduced (P-value <0.001). When compared to Saline-left and untreated condyles, BoNTA-right condyles showed significant bone loss (P-value <0.001) and shape changes. No significant bone loss was observed in the alveolar processes of any of the groups (P-value >0.05). Conclusion Condyle bone quality deteriorates at an early stage of BoNTA-induced masseter muscle atrophy, and before the alveolar process is affected. Since the observed bone microstructural changes resemble those in human temporomandibular joint degenerative disorders, the clinical safety of BoNTA intervention in the masticatory apparatus remains to be clarified.

Dental wear patterns reveal dietary ecology and season of death in a historical chimpanzee population

Julia Stuhltraeger, Ellen Schulz-Kornas, Ottmar Kullmer, Marcel M. Janocha, Roman M. Wittig, Kornelius Kupczik

Summary: This study combines macroscopic and microscopic wear analyses to explore tooth use differences between historical western chimpanzees from Liberia and the extant chimpanzees of Ta1 National Park in Ivory Coast. The results indicate similar long-term feeding behavior but significant short-term differences between the populations, potentially linked to historical events during dry periods in Liberia. Furthermore, differences in 3D surface textures between Liberian males and females suggest a higher consumption of insects by females, in line with patterns observed in Ta1 chimpanzees.

The cost of chewing: The energetics and evolutionary significance of mastication in humans

Summary: Chewing different items can result in varying metabolic costs, and chewing by human subjects represents a measurable energy sink. Chewing tasteless and odorless gum increases metabolic rate, with energy expenditure increasing with gum stiffness and requiring greater muscle recruitment. This adds an previously unexplored energetic dimension to the interpretation of hominin dentofacial fossils.

Mechanical Disturbance of Osteoclasts Induces ATP Release That Leads to Protein Synthesis in Skeletal Muscle through an Akt-mTOR Signaling Pathway

Camilo Morales-Jimenez, Julian Balanta-Melo, Manuel Arias-Calderon, Nadia Hernandez, Fernan Gomez-Valenzuela, Alejandro Escobar, Enrique Jaimovich, Sonja Buvinic

Summary: The present study reveals that osteoclasts can promote skeletal muscle protein synthesis by releasing ATP. This ATP release activates a series of cellular signaling pathways that lead to protein synthesis in skeletal muscle. These findings have significant implications for the understanding and clinical implications of the interaction between bone and muscle.

Summary: In this study, the researchers investigated the morphological variation of nasal airways in different geographic populations. They found subtle but statistically significant differences in nasal shape between the populations. The French and Russian samples had longer and narrower nasal airways, while the Cambodian sample had wider and shorter nasal airways. The Chilean sample showed the greatest intra-population variation, and the South African sample overlapped with the French, Russian, and Cambodian variation. Interestingly, there was no correlation between nasal airway volume and geographic groups, raising questions about climate-related oxygen consumption demands.

Fibroblast growth factor 21 is expressed and secreted from skeletal muscle following electrical stimulation via extracellular ATP activation of the PI3K/Akt/mTOR signaling pathway

Summary: Fibroblast growth factor 21 (FGF21) is a hormone involved in the regulation of lipid, glucose, and energy metabolism. Recent studies have shown that FGF21 is also synthesized and secreted by skeletal muscles through a signaling pathway involving extracellular ATP (eATP) and the Akt/mTOR pathway. This study investigated the effect of electrical stimulation on the production and secretion of FGF21 in skeletal muscles, and found that it increased both mRNA and protein levels of FGF21 through eATP signaling and the PI3K/Akt pathway. These findings provide new insights into the regulation of FGF21 production in skeletal muscles and may lead to the identification of new targets for studying physiological and pathological conditions associated with FGF21.

Summary: FMR1 gene mutations lead to fragile X syndrome and cognitive disorders, but in mice, the absence of FMR1 gene leads to increased bone density and improved bone properties. The absence of FMR1 gene also increases osteoblast differentiation and bone formation, resulting in higher bone mass and strength.

Molar biomechanical function in South African hominins Australopithecus africanus and Paranthropus robustus

Michael A. Berthaume, Kornelius Kupczik

Summary: This study quantified the molar biomechanical performance of two Plio-Pleistocene hominins, Paranthropus robustus and Australopithecus africanus, to investigate potential dietary differences between them. The results showed that Paranthropus robustus required more force and energy to fracture food items but had a higher force transmission rate. Three evolutionary scenarios concerning the dietary ecologies of these hominins were proposed based on the findings, highlighting the importance of a holistic approach to reconstructing hominin dietary ecology.
