Summary: YAP1 is highly upregulated in peritoneal carcinomatosis tumor cells, contributing to cancer stem cell properties and metastasis. Inhibition of YAP1 can significantly slow tumor growth and eliminate peritoneal carcinomatosis in preclinical models. Pharmacological inhibition of YAP1 can reduce cancer stem cell-like properties and suppress tumor growth, especially in combination with cytotoxic drugs in vivo models.

Transcription factors, such as ZEB1/ZEB2, SNAIL, and TWIST, play important roles in driving the EMT process. This review focuses on understanding the molecular mechanisms that sustain high levels of these transcription factors in aggressive carcinomas and their upregulation by transforming growth factor-β during cancer progression.

Summary: In this study, the mutants of Colletotrichum gloeosporioides lacking hypothetical sensors of plant surface signals, Delta CgMsb2 and Delta CgMsb2Sho1, were examined. The mutations adversely affected conidial size, sporulation, and growth. Transcriptional changes were observed for a large number of genes, especially those involved in hydrolases, ABC transporters, and MAPKs. The encoded MAPKs participate in signal transduction and metabolic regulation. These findings provide new insights into the regulation of C. gloeosporioides's development and infection.

Summary: This study explores the mechanism of the response of the cAMP-PKA pathway to heat shock and thermal adaptation in yeast. The results demonstrate that heat shock leads to the assembly of TPK1 mRNA in cytoplasmic foci, and the protein level of Tpk1 significantly increases during the recovery period. The expression of TPK1 upon heat shock is dependent on the Wsc3 sensor and the CWI signaling pathway. This study reveals the specificity regulation of cAMP-PKA signaling by thermal stress.

Summary: Most adult hippocampal neural stem cells (NSCs) remain quiescent, but the activity of the transcriptional co-activator Yap1 is enriched in active NSCs. Genetic deletion of Yap1 reduces the proportion of active NSCs, indicating its role in regulating the transition from quiescence to activation. Overexpression of a gain-of-function Yap1 mutant leads to cell cycle entry in NSCs and astrocytes, and dysregulated Yap1 activity represses neurogenesis and induces glioblastoma stem cell-like characteristics.
