Summary: Antibiotic resistance is a global public health concern, and the development of novel antimicrobials is urgently needed. In this study, a series of new peptidomimetic antimicrobials were designed and synthesized, with compound 51 identified as the lead compound due to its potent antibacterial activity, low toxicity, and avoidance of bacterial resistance development. Compound 51 also significantly reduced microbial burden in a murine model of keratitis infected by Staphylococcus aureus or Pseudomonas aeruginosa. This design strategy provides a promising and effective solution to overcome antibiotic resistance.

Summary: alpha-Fluorinated chalcones were synthesized and tested for their inhibitory effects on human cancer cell growth. The most potent compound, 4c, demonstrated selective toxicity against cancer cells and disrupted microtubule networks, leading to cell cycle arrest and apoptosis. Furthermore, compound 4c inhibited angiogenesis and metastasis of cancer cells. Overall, chalcone hybrid 4c is a promising anti-cancer agent targeting tubulin assembly.

Summary: The existence of antimicrobial encrypted fragments in human proteins has been demonstrated, and algorithms have been developed to identify these segments on a large scale. These fragments are typically cationic and amphiphilic segments that exert antimicrobial activity by disrupting membranes. The study found that about 2% of all human proteins contain these segments, with a higher prevalence in transmembrane and peripheral membrane proteins. These segments have conservation scores similar to other residues with similar solvent accessibility.

Summary: Antibiotic resistance and drug-resistant bacterial infections are significant threats to public health. In this study, synthetic mimics of antimicrobial peptides based on functional poly(methionine)s were developed. These poly(methionine) compounds demonstrated potent antibacterial activity against antibiotic-resistant bacteria, as well as good biocompatibility and bactericidal performance. The findings suggest that sulfonium poly(methionine)s have the potential to address the challenge of drug-resistant bacterial infections.

In this study, cationic nano-sized hyperbranched polymers (CNHBPs) were constructed, and one particular type showed the highest antimicrobial and antibiofilm activities. It also exhibited superior therapeutic efficacy in a mouse wound model of MRSA infection.
