LAIR-1 overexpression and correlation with advanced pathological grade and immune suppressive status in oral squamous cell carcinoma

Background This study aims to investigate the characteristic role of inhibitory receptor leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1) in oral squamous cell carcinoma (OSCC). Methods The expressions of LAIR-1 and other immune-related molecules were detected in a human OSCC tissue microarray. LAIR-1 expression difference among different clinicopathological parameters was analyzed. The correlations of LAIR-1 with several immune-related markers were assessed. Results Compared with dysplasia and oral mucosa, the expression of LAIR-1 was significantly upregulated in the stroma of OSCC, and its overexpression was correlated with advanced pathological grade. Overexpression of LAIR-1 was significantly associated with tumor-associated macrophage and myeloid-derived suppressor cell markers (CD68, CD163; CD33, CD11b), indoleamine 2,3-dioxygenase (IDO) and two immune checkpoints (B7-H3 and VISTA). Conclusions Overexpression of LAIR-1 was associated with advanced pathological grade and correlated with immune suppressive features in OSCC. Further studies are required to identify the specific immunological role of LAIR-1.

Ferroptosis promotes anti-tumor immune response by inducing immunogenic exposure in HNSCC

A Dual-Responsive STAT3 Inhibitor Nanoprodrug Combined with Oncolytic Virus Elicits Synergistic Antitumor Immune Responses by Igniting Pyroptosis

Summary: This study reports a combinational therapeutic strategy that combines tumor microenvironment-reactive oxygen species/pH dual-responsive signal transducers and activators of transcription 3 inhibitor nanoprodrugs with oncolytic herpes simplex virus 1 virotherapy to enhance immunotherapy. This strategy successfully activates robust T-cell-dependent antitumor responses, potentiating immune checkpoint blockade effects against local recurrence and pulmonary metastasis in preclinical cold murine triple-negative breast cancer and syngeneic oral cancer models.

The rapid advancement of advanced materials and technologies has also revolutionized the treatment methods and strategies for oral diseases. This review provides a systematic summary of the application strategies in advanced materials and technologies, based on the etiology of the diseases and a comparison of old and new materials, as well as the challenges and future directions for their development in oral disease treatment. It will serve as a guide for both fundamental research and clinical translation in oral medicine.

Diselenide-Based Dual-Responsive Prodrug as Pyroptosis Inducer Potentiates Cancer Immunotherapy

It can remodel the immunostimulated tumor microenvironment and trigger a robust immune response. Combined with αPD-1 therapy, it effectively inhibits the growth of remote tumors and prolongs survival.

Biomimetic manganese-based theranostic nanoplatform for cancer multimodal imaging and twofold immunotherapy

Summary: We have developed a nanoplatform capable of improving the immunosuppressive tumor microenvironment (TME) by actively targeting it. By camouflaging the surface of gold nanorods (GNRs) with myeloid-derived suppressor cells (MDSCs) membrane, we have achieved targeted adjustment of the TME. This nanoplatform also enables multimodal cancer imaging and dual immunotherapy.

Summary: This study investigated the spatial architecture of immune cells in tertiary lymphoid structures (TLSs) associated with head and neck squamous cell carcinoma (HNSC). It identified chemokine receptors associated with TLSs and portrayed the cellular atlas using multiplex immunohistochemistry. The study also explored the functions and evolutionary trajectory of cells in TLSs, and proposed a method to enhance the antitumor immune response by inducing neogenesis TLSs in HNSC.

Tumor-host colluding through erythroid progenitor cells: Mechanisms and opportunities

The findings suggest the potential for further research on EPCs and their mechanisms in tumor treatment.

Tertiary Lymphoid Structure Raises Survival and Immunotherapy in HPV- HNSCC

Summary: Immune checkpoint blockade therapy targeting PD-1/PD-L1 shows limited efficacy in head and neck squamous cell carcinoma (HNSCC), however, recent findings suggest that the presence of tertiary lymphoid structures (TLSs) in tumor tissue is associated with better prognosis and response to therapy. This study analyzed TCGA-HNSCC data and identified an immune classification for the tumor microenvironment (TME) of HNSCC and found that TLS enrichment was correlated with improved prognosis and response to immune checkpoint blockade treatment. Further experiments demonstrated that inducing TLS formation enhanced the response to PD-1 blockade treatment in HPV- HNSCC mouse models by increasing the presence of specific immune cells in the TME. The results suggest that inducing TLS formation could be a potential therapeutic approach for improving the response rate to immune checkpoint blockade therapy in HPV- HNSCC patients.

Innovative Strategies for Hair Regrowth and Skin Visualization

Summary: Around 50% of men and 15-30% of women suffer from hair-related problems, causing significant psychological burden. Traditional treatments, such as drug therapy and transplantation, face challenges like adverse effects and poor drug penetration. Therefore, efforts have been made to enhance drug permeation for hair regrowth. This review focuses on transdermal strategies involving external stimulation, regeneration, and microneedles, as well as natural products as alternative agents. It also discusses skin visualization strategies and provides information on relevant patents and clinical trials.

Targeting PCSK9 reduces cancer cell stemness and enhances antitumor immunity in head and neck cancer

PCSK9 inhibition enhances CD8(+) T cell infiltration, reduces MDSCs, and improves the efficacy of anti-PD-1 ICB therapy in HNSCC.

CSRP2 promotes cell stemness in head and neck squamous cell carcinoma

Activation of Pyroptosis Using AIEgen-Based sp(2) Carbon-Linked Covalent Organic Frameworks

Summary: Covalent organic frameworks (COFs) have shown potential as porous materials for cancer phototherapy due to their unique characteristics. However, the aggregation-caused quenching effect and apoptosis resistance limit their effectiveness. This study demonstrated that linking AIEgens into COF networks was an effective strategy for inducing pyroptosis and enhancing antitumor immunity. The synergized system of AIE COF and αPD-1 effectively eradicated tumors and inhibited recurrence and metastasis in a tumor model.

Summary: Cancer stem cell-like cells (CSCs) play a crucial role in the heterogeneity, metastasis, and treatment resistance of head and neck squamous cell carcinoma (HNSCC). The gene LIMP-2 has been identified as a potential therapeutic target for regulating HNSCC progression and CSC properties. High expression of LIMP-2 suggests poor prognosis and potential immunotherapy resistance in HNSCC patients. Mechanistically, enhanced autophagy mediated by LIMP-2 helps HNSCC maintain stemness and facilitates the transcription of downstream target genes.
