Adipose tissue-derived mesenchymal stem cells cultured at high density express IFN-β and TRAIL and suppress the growth of H460 human lung cancer cells

Although mesenchymal stem cells (MSCs) have been reported to inhibit tumor growth, the mechanism controlling this tumor suppression function is unclear. Here, we report that high-density (40,000 cells/cm(2)) cultured adipose tissue-derived MSCs (40K-ASCs) expressed interferon (IFN)-beta and tumor necrosis factor-related apoptosis-inducing ligand (TRAIL); we also found that serum deprivation during cell culture induced the expression of IFN-beta and TRAIL. In addition, the mRNA expression of IFN-beta, but not TRAIL, was increased during the washing step required for the transplantation of normal-density (5000 cells/cm(2)) cultured ASCs (5K-ASCs). When the human lung cancer cell line H460 was co-cultured with 40K-ASCs, necrotic cell death was dramatically increased in vitro. When ASCs were injected after four washes, both 5K-ASCs and 40K-ASCs substantially reduced tumor weight in H460-derived cancer animal models. These results suggest that serum deprivation during the culture of 40K-ASCs or during the washing step of 5K-ASCs can induce IFN-beta and/or TRAIL expression, ultimately leading to the tumor suppression capability of ASCs.

Characteristics of pediatric thoracic trauma: in view of before and after the establishment of a regional trauma center

Summary: Pediatric thoracic trauma differs from adults due to small anatomy and rapid recovery. Data from 168 patients under 15 years old with thoracic trauma were analyzed, with pedestrian traffic accidents being the most common cause of injury and lung contusion being the most common injury. Independent risk factors of in-hospital mortality were hemopneumothorax and cardiac contusion.

Summary: Cultured human skeletal-muscle satellite cells (SkMSCs) have shown anti-inflammatory properties by secreting prostaglandin E2 and hepatocyte growth factor (HGF). In this study, it was found that a single treatment with SkMSCs could ameliorate acute liver and gut inflammation induced by binge ethanol administration in mice, reducing markers of liver injury, bacterial-derived lipopolysaccharide levels, tissue inflammation, and leaky gut indicators.

Pulsed Electromagnetic Field (PEMF) Treatment Reduces Lipopolysaccharide-Induced Septic Shock in Mice

Summary: This study examined the effect of pulsed electromagnetic field (PEMF) therapy on a mouse model of lipopolysaccharide (LPS)-induced septic shock. The results showed that PEMF treatment increased the survival rate of mice and decreased pro-inflammatory cytokine gene expression and liver damage. Microarray analysis identified several genes regulated by PEMF therapy. Therefore, PEMF may be beneficial for patients with bacteria-induced sepsis and reduce septic shock-induced mortality.

Analysis of Missed Skeletal Injuries Detected Using Whole-Body Bone Scan Applied to Trauma Patients: A Case-Control Study

Method to Determine the Optimal Aptamer-to-Bead Ratio by Using Flow Cytometry

Summary: Research on the effective attachment of aptamers to beads has made little progress. This study demonstrates a new method using flow cytometry to determine the optimal aptamer-to-bead ratio for immobilization. The proposed method shows increased fluorescence intensity with increasing aptamer concentration, peaking at a ratio of 2.56 x 10(5), and decreasing at higher ratios. Analysis using fluorescence microscopy yielded similar results. However, there was a significant loss of aptamer compared to a ratio of 1.28 x 10(5) after the conjugation reaction, and the binding efficiency was highest at the ratio of 1.28 x 10(5). These findings suggest that the proposed method is the best and easiest for determining the optimal aptamer-to-bead ratio.
