摘要

Colorectal cancer (CRC) is one of the major causes of mortality and morbidity, and is the third most common cancer in men and the second most common cancer in women worldwide. The incidence of CRC shows considerable variation among racially or ethnically defined populations in multiracial/ethnic countries. The tumorigenesis of CRC is either because of the chromosomal instability (CIN) or microsatellite instability (MIN) or involving various proto-oncogenes, tumor-suppressor genes, and also epigenetic changes in the DNA. In this review I have focused on the mutations and polymorphisms of various important genes of the CIN and MIN pathways which have been implicated in the development of CRC.

Summary: The analysis of copy number patterns of cancer genomes assists in developing a machine learning algorithm for accurately predicting homologous recombination deficiency (HRD) status. HRD renders cancer cells vulnerable to unrepaired double-strand breaks and is an important therapeutic target. Copy number alteration (CNA) can be extracted from various data sources and used for cost-effective HRD prediction. A gradient boosting machine model (HRDCNA) was built based on CNA features, identifying specific gene inactivation and providing a robust tool for HRD prediction.

New insights on familial colorectal cancer type X syndrome

Summary: Family colorectal cancer type X (FCCTX) is a heterogeneous colorectal cancer predisposition syndrome with unknown genetic etiology. This study conducted germline exome sequencing on cancer-affected patients from families at risk for FCCTX, and identified rare and potentially pathogenic variants in known hereditary cancer genes, putative FCCTX candidate genes, and other cancer-related genes. These findings provide important clues for understanding the genetic basis of FCCTX.

A specific upregulated long noncoding RNA in colorectal cancer promotes cancer progression

Summary: This study identified a specific upregulated lncRNA (SURC) in colorectal cancer (CRC), and its high expression was associated with poorer prognosis in CRC patients. Further investigation revealed that mutated adenomatous polyposis coli (APC) genes can promote the transcription of SURC by reducing the degradation of beta-catenin protein. Functional assays showed that knockdown of SURC impaired CRC cell proliferation and tumor growth. Additionally, SURC promotes CCND2 expression by inhibiting the expression of miR-185-5p in CRC cells.

Whole Exome Sequencing reveals clinically important pathogenic mutations in DNA repair genes across lung cancer patients

Summary: By performing Whole Exome Sequencing (WES) on Pakistani lung cancer patients, this study identified clinically important pathogenic mutations in 6 DNA repair genes and revealed their potential implications on prognosis. Down-regulation of these mutated genes and hypermethylation in their promoters were observed, indicating a potential convergence of genetic and epigenetic factors driving genomic instability in lung cancer cells. These findings contribute to our understanding of lung cancer susceptibility and offer potential avenues for targeted therapeutic interventions in Pakistani patients.

Emerging Tests for Noninvasive Colorectal Cancer Screening

Summary: Colorectal cancer is a common cancer globally and is a leading cause of cancer-related deaths. Current screening methods for colorectal cancer have limitations and many people are not compliant with screening recommendations. Novel screening tests based on advances in molecular biology, genetics, and epigenetics, as well as sequencing technologies and computational analytic methods, have been developed to improve early detection of colorectal cancer.

Integrating exome sequences with electronic health records can help identify novel gene-disease relationships and pathogenic loss-of-function variants across the human genome.

A Bayesian hierarchical model to estimate DNA methylation conservation in colorectal tumors

Summary: Globally, 200 million people suffer from toxic manifestations of fluoride, including dental and skeletal fluorosis, with no currently available treatment. A study on mice exposed to environmentally relevant concentrations of fluoride revealed that fluoride may adversely affect collagen, leading to skeletal fluorosis. The study also found epigenetic regulation and changes in gene expression associated with impaired collagen synthesis and mineralization.

By studying ectopically integrated CpG islands in colorectal cancer cells, researchers found that these abnormally methylated CpG islands exhibit low levels of de novo DNA methylation, with the de novo DNA methylation activity primarily targeting CpG islands marked by the histone modification H3K36me3.

The Risk of Developing Lymphoma among Autoimmune Thyroid Disorder Patients: A Cross-Section Study

Summary: This study assessed the prevalence and association of lymphoma in patients with autoimmune thyroid diseases. The study found a 7% prevalence of lymphoma, with most patients developing non-Hodgkin lymphoma (NHL) and diffuse large B-cell lymphoma (DLBCL) being the most common subtype. The incidence of lymphoma was lower than previously reported, and all lymphoma patients had a history of Hashimoto's thyroiditis. Further national studies are needed to explore the relationship between these two diseases.

Furthermore, a significant relationship between serum 25(OH)D levels and the number of medications taken by the participants was identified.

Incidence of Inborn Errors of Metabolism in Newborn Infants: Five Years' Single-Center Experience, Jeddah, Saudi Arabia

Summary: This retrospective study aimed to determine the incidence and distribution of inborn errors of metabolism (IEMs) in newborn infants and evaluate its outcome. Among 16,494 newborn infants screened, 29 were diagnosed with IEMs, with an incidence of 1 in 569 live births and a cumulative incidence of 176 per 100,000 live births. The study identified 11 different types of IEMs, with endocrinopathies being the most common category followed by carbohydrate disorders, vitamin-responsive disorders, organic acid defects, and ketogenesis and ketolysis defects. The study highlights the importance of educating the general population about the dangers of consanguineous marriages to significantly reduce related disorders, particularly in families with a history of IEMs.

Knowledge, attitudes, and perception toward human papillomavirus among health profession students: A cross-sectional study in King Saud Bin Abdulaziz University for Health Sciences

Summary: This cross-sectional study investigated the knowledge, attitude, and perception of Health Professions Students (HPS) towards human papillomavirus (HPV) at a university in Saudi Arabia. The results showed that the HPS had low knowledge and awareness about HPV, especially regarding the link between HPV infection and cervical cancer. Students from the college of medicine demonstrated higher levels of knowledge about HPV. Therefore, effective education programs and awareness campaigns should be implemented to enhance the understanding of HPV among health professions students.

Risk Factors, Clinical Manifestations and Treatment Outcomes of Colon Cancer Patients in National Guard Hospital in Jeddah, Saudi Arabia

Summary: This study evaluated the risk factors, clinical manifestations, and treatment outcomes of colon cancer patients in NGHA, Jeddah, Saudi Arabia from January 2010 to December 2020, showing that young patients (<= 50 years) had a more aggressive presentation and worse outcome in comparison to older patients (> 60 years). Further research with larger sample size and multicenter data collection is recommended to better understand the differences observed in this study.
