Summary: Tumor-derived total RNA (TdRNA) and cell lysate (TCL) are attractive alternative sources of antigens in antitumor immunotherapy. This study compared the immune responses elicited by TdRNA and TCL against breast cancer. Both TdRNA and TCL stimulated dendritic cell (DC) maturation and induced robust cellular immune responses, but TdRNA showed stronger immunoactivities in increasing T cell population through DC activation. TdRNA combined with paclitaxel (PTX) exhibited better antitumor efficacy and lower systemic toxicity compared to TCL.

Summary: DC-based immunotherapy has gained interest in anti-cancer immunotherapies. However, the immunosuppressive mechanisms in the tumor environment, such as inhibitory immune checkpoint molecules, can reduce the efficacy of DC-mediated anti-tumoral immune responses. In this study, we found that blocking CTLA-4 expression on DCs promoted their stimulatory properties and resulted in increased T cell proliferation and cytokine production.

However, some clinical trials showed that simplex whole tumor cell vaccines can only elicit weak CD8(+) T cell-mediated antitumor responses which were not enough for effective tumor elimination. Summary: Antigen selection is important for vaccines' efficacy. A comparison of two dendritic cell-based vaccination strategies showed that peptide vaccines using neoantigens predicted on the basis of in silico peptide-MHC binding affinity do not perform well compared to whole-tumor-lysate vaccines. However, prioritizing tumor-rejecting neoepitopes based on effective in vitro peptide-MHC binding affinity and peptide immunogenicity leads to more effective vaccines.
