Intrathecal administration of IGF-I by AAVrh10 improves sensory and motor deficits in a mouse model of diabetic neuropathy

Different adeno-associated virus (AAV) serotypes efficiently transduce neurons from central and peripheral nervous systems through various administration routes. Direct administration of the vectors to the cerebrospinal fluid (CSF) could be an efficient and safe strategy. Here, we show that lumbar puncture of a nonhuman AAV leads to wide and stable distribution of the vector along the spinal cord in adult mice. AAVrh10 efficiently and specifically infects neurons, both in dorsal root ganglia (60% total sensory neurons) and in the spinal cord (up to one-third of a-motor neurons). As a proof of concept, we demonstrate the efficacy of AAVrh10 in a mouse model of diabetic neuropathy, in which intrathecal delivery of the vector coding for insulin-like growth factor (IGF-I) favored the release of the therapeutic protein into the CSF through its expression by sensory and motor neurons. IGF-I-treated diabetic animals showed increased vascular endothelial growth factor expression, activation of Akt/PI3K pathway, and stimulated nerve regeneration and myelination in injured limbs. Moreover, we achieved restoration of nerve conduction velocities in both sensory and motor nerves by AAVrh10, whereas we reached only sensory nerve improvement with AAV1. Our results indicate that intrathecal injection of AAVrh10 is a promising tool to design gene therapy approaches for sensorimotor diseases.

Proteomic quantitative study of dorsal root ganglia and sciatic nerve in type 2 diabetic mice

Summary: This study characterized the proteome of neural tissues in a type 2 diabetes mouse model and identified differential protein expression profiles related to reduced glycolytic and TCA cycle metabolism, increased lipid catabolism, dysregulation of muscle-like proteins, altered cytoskeleton-related proteins, activation of inflammatory responses, and changes in glutathione metabolism and oxidative stress. These findings suggest that energy deficiency may play a key role in the neurodegeneration observed in diabetic neuropathy.

Differential toxicity profile of secreted and processed alpha-Klotho expression over mineral metabolism and bone microstructure

These findings suggest that s-KL (but not p-KL) may be a safe therapeutic strategy for exploiting KL anti-aging protective effects.

Massive Loss of Proprioceptive Ia Synapses in Rat Spinal Motoneurons after Nerve Crush Injuries in the Postnatal Period

Summary: Peripheral nerve injuries result in the retraction of Ia synapses from a-motoneurons, impairing functional recovery and correlating with injury severity and muscle reinnervation efficiency. This study analyzed VGluT1-labeled Ia synapses after nerve crush in 10-day-old rats and found a large loss of synapses similar to more severe injuries in adults. The mechanism of synapse loss in young animals may be associated with higher microglia synaptic pruning activity and results in greater losses after milder nerve injuries.

A Novel 3D-Printed/Porous Conduit with Tunable Properties to Enhance Nerve Regeneration Over the Limiting Gap Length

Animal experiments demonstrate muscle functional recovery and abundant cellularization and axon regeneration within the conduits, suggesting their potential to overcome the limitations of autograft for repairing long nerve gaps.
