Interferons not only exert a fundamental role during inflammation and immune responses but also modulate the activity of hematopoietic stem cells during homeostatic and demand-adapted hematopoiesis. Identical mechanisms regulate the homeostasis and proliferation of leukemic stem cells (LSCs). Understanding these mechanisms may lead to novel therapeutic approaches against leukemia.

Comprehensive analysis of SARS-CoV-2 receptor proteins in human respiratory tissues identifies alveolar macrophages as potential virus entry site

Summary: COVID-19 has had significant effects on global healthcare, resulting in millions of deaths. In this study, the expression of 10 known and novel virus entry molecules was investigated using immunohistochemistry. The results showed that ACE2, ASGR1, or KREMEN1 was expressed in the majority of lung tissues, indicating their potential role in SARS-CoV-2 entry. Furthermore, the expression of these molecules was observed in alveolar macrophages, suggesting their involvement in intra-alveolar virus transmission. The study also revealed the relevance of certain molecules in different anatomical sites.

Summary: Pancreatic cancer is highly resistant to traditional therapies, posing challenges for clinical management. Researchers have identified SMARCD3 as an epigenetic regulator that plays a role in pancreatic cancer stem cells. SMARCD3, although often considered a tumor suppressor, is amplified in cancer and enriched in pancreatic cancer stem cells. Deletion of Smarcd3 improves survival in established tumors, particularly when combined with chemotherapy, and it acts in conjunction with FOXA1 to regulate lipid metabolism, which is associated with therapy resistance and poor prognosis in cancer.

Summary: Cusatuzumab, a high-affinity anti-CD70 monoclonal antibody, has shown preliminary efficacy and tolerability when combined with azacitidine in patients with newly diagnosed AML ineligible for intensive chemotherapy. The recommended phase II dose of cusatuzumab was determined to be 10 mg/kg. Objective responses were achieved in 50% of patients, with a median duration of response of 4.5 months and a median overall survival of 11.5 months. The most common treatment-emergent adverse events were infections and hematologic toxicities. Further investigation of cusatuzumab combined with current standard-of-care therapy is ongoing.

Summary: Malignant pleural mesothelioma (MPM) is a lethal malignancy caused by asbestos exposure. In this study, we found that the hyperactive MAPK pathway is a molecular driver of MPM, and blocking this pathway induces cell apoptosis. Additionally, the MAPK pathway cooperates with PARP1 to regulate ROS levels, and combining MEK inhibitors with PARP inhibitors enhances cell apoptosis.

Summary: This study aimed to explore the interplay between leukocyte activation and pulmonary endothelium in different disease stages of fatal COVID-19. The results showed that early leukocyte recruitment and secretion of proinflammatory cytokines and hypercoagulability were observed in critically ill patients infected with SARS-CoV-2. In addition, compared with control groups, COVID-19 patients exhibited increased expression of leukocyte trafficking molecules and endothelial activation.

Summary: The intestine is a complex organ with various functions including digestion, immune surveillance, and maintaining symbiotic relationships with microbiota. It has differences in structure and function along its over nine-meter length. Through multiplexed imaging and single-cell assays, this study reveals substantial differences in cell compositions across different regions of the intestine, highlighting distinct immunological niches. The findings also suggest a regulatory differentiation cascade and associate specific cell types with intestinal disease heritability. This research provides an important reference map for understanding human biology and disease.

Case Report: Paraneoplastic psoriasis in thymic carcinoma

Summary: Thymic carcinomas are rare and aggressive malignancies of the anterior mediastinum, and rarely exhibit paraneoplastic syndromes. In this case report, we describe a 36-year-old patient with progressive thymic carcinoma and paraneoplastic psoriasis occurring ten months before the initial diagnosis. Psoriatic flares during the course of the disease indicated relapse or progression of the carcinoma, making this the first reported case of paraneoplastic psoriasis in thymic carcinoma.

S3-CIMA: Supervised spatial single-cell image analysis for identifying disease-associated cell-type compositions in tissue
