Animal models of inflammatory bowel disease: a review

Inflammatory bowel disease (IBD) represents a group of idiopathic chronic inflammatory intestinal conditions associated with various areas of the GI tract, including two types of inflammatory conditions, i.e., ulcerative colitis (UC) and Crohn's disease (CD). Both UC and CD are chronic inflammatory disorders of the intestine; in UC, inflammation starts in the rectum and generally extends proximally in a continuous manner through the entire colon. Bloody diarrhea, presence of blood and mucus mixed with stool, accompanied by lower abdominal cramping, are the characteristic symptoms of the disease. While in CD, inflammatory condition may affect any part of the GI tract from mouth to anus. It mainly causes abdominal pain, diarrhea, vomiting and weight loss. Although the basic etiology of IBD is unknown, there are several factors that may contribute to the pathogenesis of this disease, such as dysregulation of immune system or commensal bacteria, oxidative stress and inflammatory mediators. In order to understand these different etiological factors, a number of experimental models are available in the scientific research, including chemical-induced, spontaneous, genetically engineered and transgenic models. These models represent a major source of information about biological systems and are clinically relevant to the human IBD. Since there is less collective data available in one single article discussing about all these models, in this review an effort is made to study the outline of pathophysiology and various types of animal models used in the research study of IBD and other disease-related complications.

Animal models of attention-deficit hyperactivity disorder (ADHD)

Summary: ADHD is a heterogeneous neuropsychiatric disorder characterized by symptoms of hyperactivity, attention deficit, and impulsiveness, which can significantly impact academic, occupational, and social functioning. Currently, there are limited effective medications available to completely eliminate ADHD symptoms, making it essential to understand the neurochemical and genetic alterations that contribute to the progression of the disorder. Animal models serve as a valuable tool for studying the pathological mechanisms involved in disease progression and have good predictive validity for the development of new therapeutic interventions.

Berberine Ameliorate Haloperidol and 3-Nitropropionic Acid-Induced Neurotoxicity in Rats

Summary: Movement disorders are neurological conditions that cause involuntary motor movements. Excessive ROS generation, mitochondrial dysfunction, neuroinflammation, and neurotransmitter imbalance contribute to the development of motor dysfunction. The Nrf2/HO-1 antioxidant pathway and GSK-3 beta play important roles in neuroprotection against movement disorders.

Research suggests that GPER has the potential to slow down the progression of neurodegenerative disorders.

Opportunities and challenges in application of artificial intelligence in pharmacology

Summary: Artificial intelligence (AI) is a machine science that imitates human behavior and intelligent data analysis using specialized algorithms, deep learning, and machine learning. It provides an automated and reliable tool for accurately and rapidly making decisions in the digital world where a vast amount of medical data is generated daily. AI has been widely applied in various fields in pharmaceutical science, particularly in pharmacological research, aiding in the analysis of preclinical and clinical trial data, drug discovery/manufacturing, disease diagnosis, personalized treatment, clinical trial research, radiotherapy, surgical robotics, electronic health records, and epidemic outbreak prediction. Additionally, AI is instrumental in evaluating biomarkers and diseases. This review explains the models and processes of machine learning and discusses their role in pharmacological science, illustrating the relevance and potential of AI with deep learning and machine learning in pharmacological research.

Impact of noscapine on halting the progression of pentylenetetrazole induced kindling epilepsy in mice

Summary: Epilepsy is a disease characterized by excessive excitatory neuronal firing and motor, psychomotor, and sensory impairments. Current therapies fall short in producing satisfactory outcomes due to the complexity of the disease, inadequate diagnosis, and the rise of drug-resistant epilepsy. This study investigated the potential of repurposing the drug "noscapine" for the management of epilepsy and its associated complications. The results showed that noscapine pretreatment attenuated the behavioral and biochemical changes induced by prolonged PTZ treatment in mice, suggesting its potential as a new antiepileptic agent.

Effect of Bacille-Calmette-Guerin vaccine against rotenone-induced Parkinson's disease: Role of neuroinflammation and neurotransmitters

Summary: This study aimed to investigate the neuroprotective effect of the BCG vaccine on the rotenone-induced Parkinson's disease model. Rats were injected with rotenone and the BCG vaccine, and behavioral assessments and biochemical analysis were performed. The results showed that the BCG vaccine significantly improved motor deficits, restored biochemical levels, normalized neurotransmitter levels, and reduced inflammatory cytokines in the striatum. These findings suggest that the BCG vaccine has neuroprotective properties and could be used for other neurological disorders, including PD.

Additionally, it increased the levels of antioxidant enzymes, reduced oxidative stress and apoptotic markers, and restored neurotransmitter levels.
