Platelet miRNA Biosignature Discriminates between Dementia with Lewy Bodies and Alzheimer's Disease

Ana Gamez-Valero, Jaume Campdelacreu, Dolores Vilas, Lourdes Ispierto, Jordi Gascon-Bayarri, Ramon Rene, Ramiro Alvarez, Maria P. Armengol, Francesc E. Borras, Katrin Beyer

Summary: This study identified a seven-miRNA biosignature that can distinguish DLB from AD, and identified three disease-specific clusters of pathways as a result of platelet-miRNA deregulation.

B Cell-Derived Extracellular Vesicles Reveal Residual B Cell Activity in Kidney Graft Recipients Undergoing Pre-Transplant Desensitization

David Cucchiari, Valeria Tubita, Jordi Rovira, Maria J. Ramirez-Bajo, Elisenda Banon-Maneus, Marta Lazo-Rodriguez, Natalia Hierro-Garcia, Francesc E. Borras, Pedro Ventura-Aguiar, Gaston J. Pineiro, Jaume Martorell, Lluis Peri, Mireia Musquera, Alexandre Hertig, Federico Oppenheimer, Josep M. Campistol, Fritz Diekmann, Ignacio Revuelta

Summary: The study found that in LDKT recipients undergoing desensitization for HLA-incompatibility, BEVs significantly decreased after desensitization, and relapse occurred in patients who later developed ABMR. These findings were validated in other cohorts, confirming that BEVs could reflect B cell residual activity after desensitization and serve as a valid surrogate for humoral alloreactivity in this setting.

Local administration of porcine immunomodulatory, chemotactic and angiogenic extracellular vesicles using engineered cardiac scaffolds for myocardial infarction

Marta Monguio-Tortajada, Cristina Prat-Vidal, Miriam Moron-Font, Marta Clos-Sansalvador, Alexandra Calle, Paloma Gastelurrutia, Adriana Cserkoova, Anna Morancho, Miguel Angel Ramirez, Anna Rosell, Antoni Bayes-Genis, Carolina Galvez-Monton, Francesc E. Borras, Santiago Roura

Summary: The study presents a novel MSC-EV delivery approach for cardiac repair, showing significant effects in reducing immune response, promoting angiogenesis, and reducing inflammation. The research demonstrates the potential therapeutic efficacy of loading EV in engineered pericardial scaffolds for treating myocardial infarction.

Reduced Plasma Extracellular Vesicle CD5L Content in Patients With Acute-On-Chronic Liver Failure: Interplay With Specialized Pro-Resolving Lipid Mediators

Maria Belen Sanchez-Rodriguez, Erica Tellez, Mireia Casulleras, Francesc E. Borras, Vicente Arroyo, Joan Claria, Maria-Rosa Sarrias

Summary: This study analyzed the macrophage anti-inflammatory protein CD5L in the plasma extracellular vesicles (EVs) of patients with acute-on chronic liver failure (ACLF) and its relationship with lipid mediators. The circulating EV contents in ACLF were significantly altered, leading to a loss of anti-inflammatory and pro-resolving molecules involved in the control of acute inflammation.

Mesenchymal stromal cells induced regulatory B cells are enriched in extracellular matrix genes and IL-10 independent modulators

Sergio G. Garcia, Noelia Sandoval-Hellin, Marta Clos-Sansalvador, Laura Carreras-Planella, Miriam Moron-Font, Dolores Guerrero, Francesc E. Borras, Marcella Franquesa

Summary: This study characterized B cell populations induced by MSC and identified specific biomarkers using RNAseq. The results showed that the induced B cell populations had immunomodulatory and extracellular matrix generation capabilities independent of IL-10.

Borras, Francisco Rodriguez-Frias, Ariadna Rando, Clara Ramirez-Serra, Silvia Camos, Esteban Domingo, Marta Bes, Celia Perales, Maria Isabel Costafreda

Summary: MicroRNAs (miRNAs) encapsulated in extracellular vesicles (EVs) have potential as diagnostic and prognostic biomarkers. This study aimed to find a reliable EV isolation method for miRNA sequencing, suitable for clinical applications. Results showed that SEC yielded the highest amount of EVs with good reproducibility, while GRAD and SEC+GRAD resulted in purer EV preparations. The NEBNext library exhibited the highest reproducibility in miRNA recovery and diversity. Therefore, combining GRAD EV isolation with NEBNext library preparation is suitable for miRNA detection using plasma samples.

Summary: Urine is commonly used for clinical diagnosis and biomedical research, and the discovery of extracellular vesicles (EV) in urine has opened a new scientific field. However, methods for isolating and characterizing urinary extracellular vesicles (uEVs) need further optimization and standardization to advance scientific research and translation into clinical practice. Recommendations for improved rigor, reproducibility, and interoperability in uEV research are provided to address current challenges and gaps in the field.
