Mucosal retinoic receptor-related orphan receptor (ROR)gamma t-expressing innate lymphoid cells (ILC) play an important role in the defense against intestinal pathogens and in promoting epithelial homeostasis and adaptation, thereby effectively protecting the vertebrate host against intestinal inflammatory disorders. The functional activity of ROR gamma t(+) ILC is under the control of environmental cues. However, the molecular sensors for such environmental signals are largely unknown. Recently, the aryl hydrocarbon receptor (AhR) has emerged as a master regulator for the postnatal maintenance of intestinal ROR gamma t(+) ILC and intraepithelial lymphocytes. AhR is a highly conserved transcription factor whose activity is regulated by environmental and dietary small molecule ligands. Here, we review the role of AhR signaling for the maintenance of intestinal immune cells and its impact on the immunological protection against intestinal infections and debilitating chronic inflammatory disorders.

Summary: Tracing SARS-CoV-2 population transmission chains remains a major challenge, but this study presents an integrated system of genomic surveillance that is capable of detecting infection chains in a large city during ongoing community transmission. The system combines viral surveillance sequencing, genetically based identification of infection clusters, integration of public health authority contact tracing data, and a user-friendly dashboard application. This integrated system can contribute to the monitoring and potentially improve the management of SARS-CoV-2 transmission in the population.

Summary: Keratinocytes (KCs) are essential for protecting the body against pathogens, and deficiencies in IL-17RA can lead to spontaneous Staphylococcus aureus skin infections. This study found an expansion of T17 cells in the skin-draining lymph nodes of mice lacking IL-17RA or both IL-17A and IL-17F. Contrary to previous suggestions, this expansion was not due to a negative feedback loop but rather depended on the microbiota and KC deficiency in IL-17RA signaling.

Regulation of innate immune system function by the microbiome: Consequences for tumor immunity and cancer immunotherapy

Summary: Innate effector cells play an important role in regulating inflammation and the development of cancer. The tumor microenvironment and microbiota can modify the functions of these cells, reducing their ability to initiate anti-tumor immune responses. The microbiota can also stimulate innate cells at mucosal tissues and influence immune responses in non-mucosal tissues, impacting cancer development. Additionally, the microbiota has been found to enhance innate immune responses during anti-cancer therapies. This review discusses the role of the microbiota in shaping host innate immune responses in cancer.

Summary: It is commonly believed that surgical infections are primarily caused by environmental or cutaneous bacteria. Accordingly, efforts to prevent postoperative infections have focused on improving hygiene and asepsis. However, our study revealed that the main causative bacteria in postoperative infections are of intestinal origin, which was also confirmed in mice undergoing partial hepatectomy. We found that CCR6+ group 3 innate lymphoid cells (ILC3s) play a crucial role in limiting systemic bacterial spread, and this function is dependent on the production of interleukin-22 (IL-22) and the expression of antimicrobial peptides in hepatocytes. Our findings highlight the significance of endogenous intestinal bacteria as a source of postoperative infection and suggest that ILC3s could be potential therapeutic targets.
