Summary: A tolerogenic nanovaccine has been developed and tested on an animal model of multiple sclerosis. The nanovaccine delivers a self-antigen to dendritic cells, blocking the interaction between immune cells and reducing interferon gamma secretion. The results show that the nanovaccine has significant preventive and therapeutic effects in the experimental autoimmune encephalomyelitis (EAE) model.

Summary: GM-CSF is a glycoprotein with immunomodulatory properties derived from both hemopoietic and nonhemopoietic sources. In patients with diabetic foot ulcers, GM-CSF activation is impaired, leading to impaired inflammatory response. Correcting macrophage immune dysfunction with exogenous GM-CSF may help restore immune balance and promote wound healing.

Summary: West Nile virus (WNV) is the main cause of epidemic arboviral encephalitis in the United States. Depleting microglia in WNV-infected mice enhances viral replication and tissue injury, while augmenting microglial activation through GM-CSF administration reduces viral titers and apoptotic activity and increases survival. Stimulation of microglial activation may be a viable therapeutic approach for WNV neuroinvasive disease.

Summary: This study analyzed the efficacy and safety of granulocyte-macrophage colony-stimulating factor (GM-CSF) antibodies in COVID-19 patients. The results showed that GM-CSF antibody treatment can reduce the incidence of invasive mechanical ventilation and have slight but not significant effects on mortality and ventilation improvement. Sensitivity analysis revealed statistically significant results for mortality and ventilation improvement rate when one study was excluded. Furthermore, GM-CSF antibodies did not increase the risks of secondary infection and adverse events.

Summary: This study aims to understand the role of granulocyte-macrophage-colony-stimulating factor (GM-CSF)-producing T helper (ThGM) cells in the pathophysiology of mouse experimental autoimmune uveitis (EAU). Summary: This study reveals that the pentose phosphate pathway (PPP) regulates the clearance of tolerogenic apoptotic cells (ACs) and immune tolerance. ACs decrease the levels of PPP-related genes and metabolites in macrophages. The PPP agonist AG1 increases PPP activity but greatly reduces macrophage phagocytosis of ACs and enhances the inflammatory response during efferocytosis. MiR-323-5p regulates the expression of PPP-related genes and its levels increase during efferocytosis.

Summary: This study investigates the regulation of IL-2 signaling in CD4+ T cells by soluble CD25 (sCD25). The results show that sCD25 reversibly sequesters IL-2, limiting acute proliferative responses but maintaining IL-2 availability for prolonged culture. sCD25 inhibits T-cell growth and CD25 expression, resulting in a lower prevalence of CD25high T cells. Additionally, sCD25 competes with CD25 and IL-2 receptors for limited pools of IL-2, leading to preferential enrichment of CD25high memory/effector and regulatory FOXP3+ subsets.
