The cellular and molecular mechanisms of immuno-suppression by human type 1 regulatory T cells

The immuno-regulatory mechanisms of IL-10-producing type 1 regulatory T (Tr1) cells have been widely studied over the years. However, several recent discoveries have shed new light on the cellular and molecular mechanisms that human Tr1 cells use to control immune responses and induce tolerance. In this review we outline the well known and newly discovered regulatory properties of human Tr1 cells and provide an in-depth comparison of the known suppressor mechanisms of Tr1 cells with FOXP3(+) T-reg. We also highlight the role that Tr1 cells play in promoting and maintaining tolerance in autoimmunity, allergy, and transplantation.

Editing T cell repertoire by thymic epithelial cell-directed gene transfer abrogates risk of type 1 diabetes development

Summary: The study developed a gene transfer approach to modify the T cell repertoire in thymic epithelial cells, correcting the autoimmune response in type 1 diabetes. The strategy involved intrathymic injection of a lentiviral vector to achieve stable transgene expression in thymic epithelial cells, leading to successful correction of the disease in mice models.

IL-10-Engineered Dendritic Cells Modulate Allogeneic CD8+ T Cell Responses

Summary: Tolerogenic dendritic cells (tolDC) play a central role in immune homeostasis and peripheral tolerance. We developed a protocol to generate genetically engineered tolDC overexpressing IL-10 (DCIL-10) and found that they can modulate cytotoxic CD8(+) T cell responses by reducing proliferation and activation, and inducing anergic CD8(+) T cells without signs of exhaustion.

Aryl hydrocarbon receptor activity downstream of IL-10 signaling is required to promote regulatory functions in human dendritic cells

Summary: IL-10 induces the generation of tolerogenic dendritic cells (tolDC) from monocytes through the activation of aryl hydrocarbon receptor (AHR). AHR activity downstream of IL-10 signaling is required for the induction of tolerogenic activities in DCs, and this IL-10/AHR genomic signature is active in circulating DCs in healthy individuals. However, in multiple sclerosis patients, an altered IL-10/AHR signature is observed, which correlates with functional defects and reduced frequencies of IL-10-induced tolDCs in vitro and in vivo.
