Summary: Tolerogenic dendritic cells are generated by genetic engineering of monocytes with lentiviral vectors encoding immunodominant antigens and IL-10. These cells downregulate antigen-specific T cell responses and induce antigen-specific regulatory T cells, offering a potential platform for controlling T-cell mediated diseases. In preclinical models, the administration of these tolerogenic dendritic cells prevents the development of type 1 diabetes.

Summary: Castleman disease (CD) is a rare lymphoproliferative disorder with various clinico-pathological subtypes. The pathogenesis of CD is still uncertain, but alterations in T-cell subsets may contribute to the development of CD. In this study, the distribution of T-cell subsets in the clinico-pathological spectrum of CD was investigated. The results showed decreased CD4/CD8 ratio and lower number of FOXP3+ T-reg cells in CD cases compared to non-specific reactive lymph nodes.

Summary: IL-10 induces the generation of tolerogenic dendritic cells (tolDC) from monocytes through the activation of aryl hydrocarbon receptor (AHR). AHR activity downstream of IL-10 signaling is required for the induction of tolerogenic activities in DCs, and this IL-10/AHR genomic signature is active in circulating DCs in healthy individuals. However, in multiple sclerosis patients, an altered IL-10/AHR signature is observed, which correlates with functional defects and reduced frequencies of IL-10-induced tolDCs in vitro and in vivo.
