Antioxidant activity of polyphenolic extracts of Ichnocarpus frutescens

In the present study antioxidant activities by (1,1-diphenyl-2-picrylhydrazyl radical (DPPH), hydrogen peroxide, hydroxyl radical inhibition, hemolysis by hydrogen peroxide assay, reducing power and total antioxidant activities of polyphenolic extract of Ichnocarpus frutescens leaves were investigated. The flavonoids and total polyphenolic contents of the extract were also determined using standard methods. Phytochemical analyses revealed the presence of flavonoids, polyphenols, anthocyanins and simple phenolic acids. The results of antioxidant activities of polyphenol extract obtained by different in vitro methods were varied depending on the method used. Nevertheless, polyphenol extract showed significant inhibitory activities in all in vitro reactive oxygen species scavenging, might be attributed due to the high level of polyphenolic compound. Also, these various antioxidant activities were compared to alpha-tocopherol and L-ascorbic acid as reference antioxidant compounds. These findings provide evidence that the polyphenolic extract of I. frutescens is a natural source of antioxidant against oxidative damage. (c) 2012 King Saud University. The cardioprotective actions of tilianin are believed to be achieved through mechanisms such as free radical scavenging, inflammation control, and mitochondrial function regulation.

Summary: Kirenol, a natural diterpenoid molecule found in Sigesbeckia species, has demonstrated significant potential in reducing inflammation. This review summarizes the efficacy and safety of kirenol in treating inflammation, its potential mechanisms of action, and opportunities for future drug development. Preclinical studies have shown that kirenol has therapeutic potential against various inflammatory diseases, including multiple sclerosis, inflammatory bowel disorders, and arthritis. Molecular mechanisms contributing to kirenol's anti-inflammatory actions include the inhibition of pro-inflammatory cytokines and the stimulation of heme-oxygenase-1 expression. The review also addresses the challenges and opportunities of drug delivery for kirenol in inflammation treatment.

Summary: This study aims to develop a nanofibrous wound dressing for topical drug delivery in wound healing applications. The electrospinning technique is used to fabricate bioabsorbable nanofiber scaffolds enriched with thrombin and vancomycin. The nanofibrous scaffolds demonstrate excellent biocompatibility, antimicrobial activity, and wound healing capability in both in vitro and in vivo studies. Based on the results, it can be concluded that the nanofibrous scaffolds loaded with thrombin and vancomycin have promising potential as a dressing material for hemostatic wound healing applications.

Research Article In vitro and in vivo Modulation of Postprandial Hyperglycemia by Solanum incanum L. (Bitter Apple)

Summary: This study evaluated the hypoglycemic, α-amylase, DPP-IV, and free radical scavenging effects of a hydroalcoholic extract of Solanum incanum (HAE-SI) in various in-vitro and in-vivo models. The results showed that HAE-SI has a hypoglycemic effect on rats fed a high fructose diet and inhibits α-amylase and DPPIV, as well as exhibits antioxidant properties in in-vitro assays.

Summary: Streptonigrin, an aminoquinone alkaloid derived from Streptomyces flocculus, has potential antitumor and antibiotic effects. This comprehensive review aims to discuss the prospects of streptonigrin and its derived compounds as interesting target molecules for new drug design, development, and therapy, providing a reference for their development and utilization as pharmaceuticals.

Synthesis, pharmacological evaluation, and molecular modeling studies of novel isatin hybrids as potential anticancer agents

Summary: A series of novel isatin hybrids 5a-g were designed, synthesized, and characterized spectroscopically. The synthesized compounds were evaluated for their cytotoxic activity against the human breast cancer cell line (MCF-7) using in vitro MTT assay. Compound 5e, bearing benzyl moiety at N4 piperazine, exhibited the most potent anticancer activity with an IC50 of 12.47 mM. Additionally, compounds 5e and 5g were evaluated for their antitumor activity against Dalton's ascitic lymphoma (DAL) cell line, and compound 5e showed the ability to normalize the blood picture compared to the standard drug. A molecular docking study using the crystal structure of Hsp90 protein provided insights into the protein-ligand interactions and binding mode. The drug-likeliness of the compounds was predicted based on Lipinski's rule of five and pharmacokinetic ADME parameters. Therefore, the synthesized isatin hybrids could serve as promising lead compounds for anticancer drug discovery due to their drug-like properties.

Therapeutic implications of current Janus kinase inhibitors as anti-COVID agents: A review

Summary: Severe cases of COVID-19 are characterized by hyperinflammation and multiorgan failure. JAK-STAT signaling plays a role in the immunopathogenesis of COVID-19. Jakinibs, small molecules that target proinflammatory cytokines, are being investigated as potential treatments through clinical trials. Baricitinib is currently the only FDA-approved Jakinib for treating critical COVID-19 patients. Further studies are needed to understand the pathogenesis of COVID-19 and determine the optimal duration and combination therapy of Jakinibs. This review article provides important insights into the therapeutic implications of Jakinibs for COVID-19 treatment.

Identification of BACE-1 Inhibitors against Alzheimer's Disease through E-Pharmacophore-Based Virtual Screening and Molecular Dynamics Simulation Studies: An Insilco Approach

Summary: In this study, researchers used E-pharmacophore modeling to screen the eMolecules database and identified potential drug candidates against Alzheimer's disease with enhanced physicochemical and pharmacokinetic properties. Molecular dynamics simulation and MMPBSA analysis revealed the stability and binding free energy of these compounds. Furthermore, Qikprop analysis indicated that the selected compounds have good drug-likeness and pharmacokinetic properties.

Development of a Polyherbal Topical Gel for the Treatment of Acne

Summary: The present study aimed to develop and evaluate a polyherbal gel for treating acne by formulating a mixture of Aloe barbadensis pulp and Vigna radiata extract using Carbopol 940, triethanolamine, and propylene glycol as gelling, viscosity modifying, and pH modifying agents respectively. The developed gel exhibited significant antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans, with promising inhibition zones. The gel formulation also showed beneficial effects on acne, with the additional benefit of Aloe barbadensis on the skin.

In vivo assessment of black seed oil single dose on prednisolone pharmacokinetics
