Teladorsagia circumcincta: Molecular characterisation of the avr-14B subunit and its relatively minor role in ivermectin resistance

Individual mutations (e. g. L256F) and polymorphisms in the avr-14B gene, a glutamate-gated chloride channel subunit, have been associated with ivermectin (IVM) resistance in Caenorhabditis elegans and Cooperia oncophora. The aim of the present study was to determine the full-length coding sequence of the avr-14B subunit homologue in Teladorsagia circumcincta and determine the presence/absence of the putative L256F SNP or any other potential SNPs of interest. Subsequently, we investigated sequence polymorphisms and transcription patterns between four different T. circumcincta isolates: two from Scotland (MTci1 susceptible and MTci5 triple resistant to benzimidazoles, levamisole and IVM) and two from Spain (S-Sp susceptible and R-Sp double resistant to levamisole and IVM). The complete amino acid sequence of the T. circumcincta avr-14B subunit comprises 438 amino acids. Pyrosequencing analysis failed to detect the presence of the L256F mutation in any of the MTci5 or Sp-R samples tested. However, we revealed significant allele frequency changes by means of SSCP analysis of a 106 bp region encompassing the L256F SNP. Allele E showed the greatest change, following IVM exposure in vitro and in vivo, although sequence analysis did not reveal any coding changes. Sequence analysis of the full-length avr-14B coding sequence showed that two SNPs exclusively found in the resistant strain McTi5 (I270F and T305A) are situated in codons involved in the interaction of the receptor with IVM. Moreover, other potentially significant SNPs (K361E and L364M) were identified between transmembrane regions 3 and 4. However, due to the low frequency of all these SNPs, we cannot conclude they confer IVM resistance in T. circumcincta. Moreover, a modest increase in expression of the avr-14B in both resistant isolates has been shown although these differences were not sufficiently great to consider avr-14B to be the sole or even a major determinant of IVM resistance in this species. Novel compound shows in vivo anthelmintic activity in gerbils and sheep infected by Haemonchus contortus

Summary: The control of gastrointestinal nematodes in livestock is becoming increasingly difficult due to limited drug options and the development of resistance. In this study, the potential toxicity and anthelmintic efficacy of three compounds were tested in mice and Mongolian gerbils infected with Haemonchus contortus. The benzimidazole derivative, 2aBZ, showed promising results in reducing the percentage of pre-adults in gerbil stomachs and the number of eggs shed in sheep feces. Overall, 2aBZ could be considered a promising candidate for the treatment of helminth infections in small ruminants.

Application of quantitative proteomics to discover biomarkers for tick resistance in cattle

Summary: Quantitative proteomics was used to analyze the differential abundance of serum and skin proteins in tick-resistant and tick-susceptible Brangus cattle. The study found that tick-resistant cattle had higher levels of proteins associated with immune response, blood coagulation, and wound healing compared to tick-susceptible cattle. These proteins may play a crucial role in protecting against tick infestation by promoting immune response and wound healing.

Evaluation of an indwelling bolus equipped with a triaxial accelerometer for the characterisation of the diurnal pattern of bovine reticuloruminal contractions

Summary: This observational study aimed to describe the diurnal pattern of reticuloruminal contraction rate (RRCR) and the proportion of time spent ruminating by cattle, using two commercial devices equipped with triaxial accelerometers: an indwelling bolus (placed in the reticulum) and a neck collar. The three objectives of this study were firstly to determine whether the indwelling bolus provided observations consistent with RRCR as determined by clinical examination using auscultation and ultrasound, secondly to compare estimates of time spent ruminating using the indwelling bolus and a collar-based accelerometer, and finally to describe the diurnal pattern of RRCR using the indwelling bolus data.

Further Investigations of Nitroheterocyclic Compounds as Potential Antikinetoplastid Drug Candidates

Summary: Due to the lack of specific vaccines, neglected tropical diseases caused by trypanosomatids, such as sleeping sickness, Chagas disease and leishmaniasis, rely solely on pharmacological treatments. The current drugs have drawbacks including adverse effects, parenteral administration, chemical instability, and high costs. However, the discovery of new drugs for these diseases is rare due to the lack of interest from big pharmaceutical companies. Nitroheterocyclic compounds, such as benznidazole and nifurtimox, have provided potent effects against Chagas disease, and fexinidazole has been developed as a new drug against African trypanosomiasis. Despite being initially discarded due to their mutagenic potential, nitroheterocycles now show promise as a source of inspiration for developing oral drugs to replace existing ones.

Occurrence of Leishmaniasis in Iberian Wolves in Northwestern Spain

Summary: Canine leishmaniasis is a major protozoan disease in dogs, caused by Leishmania infantum, which leads to severe health deterioration. The disease is highly prevalent in Spain, especially in coastal regions, but its presence has been spreading to other areas and cases have been reported in wildlife. This study documents the presence of wolves infected with leishmaniasis in the Sierra de la Culebra in northwestern Spain using PCR amplification of L. infantum DNA from various non-invasive samples. The positivity rate was found to be 46.1% regardless of the origin of the samples.

Quantitative TMT-based proteomics revealing host, dietary and microbial proteins in bovine faeces including barley serpin Z4, a prominent component in the head of beer

Summary: This study aimed to assess the proteome of bovine faeces and determine the origin of its component proteins, as well as the effect of treating barley with ammonia or sodium propionate preservative. Proteomic analysis identified 281 bovine proteins, 199 barley proteins, 176 bacterial proteins, and 190 archaeal proteins in the faeces. Bovine proteins included mucosal pentraxin, albumin, and digestive enzymes, while the most abundant barley protein was serpin Z4. Different bacterial and archaeal proteins were also identified. Significant differences in protein abundance were observed between the groups.

Secretory IgA as Biomarker for Gastrointestinal Nematodes Natural Infection in Different Breed Sheep

Summary: The goal of this study was to characterize IgA levels in naturally infected sheep of different breeds and explore their association with infection risk, age, and breed. Results showed that age was the determinant factor for infection risk, while breed had no impact. In addition, nasal secretions exhibited high sensitivity in detecting infection, especially in young animals.

A large deletion encompassing exon 2 of the ectodysplasin A (EDA) gene in a British blue crossbred calf with hypohidrotic ectodermal dysplasia

Summary: This study reports a case of hypohidrotic ectodermal dysplasia (HED) in cattle, in which a novel variant in the EDA gene was identified using whole-genome sequencing. The findings suggest that this variant is the genetic cause for the observed phenotype. This case highlights the potential of using whole-genome sequencing for precise diagnostics in livestock species such as cattle.
