The role of codon selection in regulation of translation efficiency deduced from synthetic libraries

Background: Translation efficiency is affected by a diversity of parameters, including secondary structure of the transcript and its codon usage. Here we examine the effects of codon usage on translation efficiency by re-analysis of previously constructed synthetic expression libraries in Escherichia coli. Results: We define the region in a gene that takes the longest time to translate as the bottleneck. We found that localization of the bottleneck at the beginning of a transcript promoted a high level of expression, especially if the computed dwell time of the ribosome within this region was sufficiently long. The location and translation time of the bottleneck were not correlated with the cost of expression, approximated by the fitness of the host cell, yet utilization of specific codons was. Particularly, enhanced usage of the codons UCA and CAU was correlated with increased cost of production, potentially due to sequestration of their corresponding rare tRNAs. Conclusions: The distribution of codons along the genes appears to affect translation efficiency, consistent with analysis of natural genes. This study demonstrates how synthetic biology complements bioinformatics by providing a set-up for well controlled experiments in biology.

Summary: A genome-wide association study identified significant genetic loci associated with suicide attempts, particularly one locus on chromosome 7 which is implicated in risk-taking behavior, smoking, and insomnia. The study also revealed strong genetic correlations between suicide attempts and psychiatric disorders, as well as other nonpsychiatric traits like smoking, pain, and lower socioeconomic status. Conditioning on psychiatric disorders reduced the genetic correlations between suicide attempts and psychiatric disorders, indicating a shared biology with nonpsychiatric risk factors.

While no significant gene main effects were found, Met allele carriers showed differences in specific brain regions compared to Val/Val individuals, suggesting a potential role of BDNF rs6265 in youth bipolar disorder. Further research is needed to explore the mediating effects of BDNF protein levels and developmental changes related to rs6265.

Summary: In this study, the association between six ABCB1 single-nucleotide polymorphisms (SNPs) and antidepressant treatment outcomes was investigated through a systematic review and meta-analysis. No significant association was found between ABCB1 SNPs and antidepressant treatment outcomes in the CAN-BIND-1 sample. However, a significant association between rs1128503 and treatment response was observed in the meta-analysis. These findings provide limited evidence for the implementation of ABCB1 genotyping to inform antidepressant treatment.

Summary: This study investigated the association between polygenic risk scores (PRS) for various psychiatric and metabolic traits and antipsychotic-induced weight gain (AIWG). The results showed nominal associations between PRSs for BMI, CAD, LDL-C, type 1 diabetes, and SCZ with AIWG. Although these associations became non-significant after multiple testing correction, these preliminary findings suggest that PRS analyses may help identify risk factors for AIWG and elucidate the underlying mechanisms.

Priming effects of a slot machine game and amphetamine on probabilistic risk-taking in people with gambling disorder and healthy controls

Summary: This study aimed to investigate the effects of gambling-specific priming manipulations and the pharmacological basis on risk-taking in a Game of Dice Task (GDT). The results showed that both gambling and AMPH increased risk-taking, with a stronger effect observed in individuals with gambling disorder. The outcomes varied with pre-treatment, and cognitive inflexibility and symptom severity moderated the effects.

Mu opioid receptor gene variant modulates subjective response to smoked cannabis

This variability in OPRM1 contributes to individual responses to cannabis and may affect risk of cannabis use disorder, highlighting potential implications for treatment targeting the endogenous opioid system.
