Summary: The study reveals that perturbation of the actin cytoskeleton is not sufficient to induce NF-kappa B activation and IL-6 secretion, but it reduces IL-6-induced activation of STAT3 in Hep3B cells. Additionally, IL-6 receptor proteolysis by ADAM10 is not dependent on the integrity of the actin cytoskeleton.

Summary: Five aromatic ergosterols with unique ring systems, named spectasterols A-E (1-5), were isolated from Aspergillus spectabilis. Compound 3 demonstrated cytotoxic activity and anti-inflammatory effects, inducing cell cycle arrest and apoptosis in HL60 cells, and inhibiting COX-2 levels and NF-kappa B p65 nuclear translocation.

Summary: The study showed that TRIM38 can alleviate IL-1 beta-induced apoptosis and degeneration of chondrocytes by suppressing the NF-kappa B signaling pathway. This work suggests a potential role of TRIM38 in osteoarthritis and proposes it as a new therapeutic target for the disease.

Summary: This study found that omaveloxolone exerts antioxidative, anti-inflammatory, antiapoptotic and anti-ECM degradation effects via activation of the Nrf2/ARE signaling pathway and inhibition of the NF-kappa B signaling pathway in chondrocytes in vitro and attenuates osteoarthritis progression in vivo, suggesting its potential as a therapeutic agent for OA.

Summary: This study highlights the essential role of endogenous IL-2 in immune checkpoint blockade-based immunotherapy and reveals the correlation between insufficient IL-2 signaling and T-cell exhaustion during tumor progression. The study also demonstrates that engineered mesenchymal stem cells (MSCs) delivering IL-2 can overcome ICB resistance and elicit anti-tumor immunity without toxicity. Engineered MSCs rejuvenate CD8(+) tumor-infiltrating lymphocytes (TILs) and potentiate the effects of ICB and chemotherapy.

Summary: In this study, the GP130-targeting molecule LMT-28 was found to effectively reduce lipopolysaccharide-induced bone resorption around implants in diabetic rats. LMT-28 downregulated the expression of RANKL and decreased osteoclast activity through the JAK2/STAT3 signaling pathway. These findings highlight the potential of LMT-28 as a therapeutic agent for peri-implantitis in diabetic patients.

Summary: The methyltransferase Smyd1 plays a crucial role in regulating IL-6 expression and secretion in endothelial cells by increasing NF-kappa B activity and promoting H3K4 trimethylation within the IL-6 promoter in response to LPS stimulation.

Summary: This study explored the protective mechanisms of Trans-cinnamaldehyde (TCA) on complete Freund's adjuvant (CFA)-induced arthritis. TCA can slow down the progression of arthritis and inhibit the immune/inflammatory responses via NF-kappa B modulation, with similar effects to methotrexate.

Summary: The extensive use of antibiotics has led to their presence in the environment, causing adverse effects on organisms. This study quantified transgenerational immune disturbances in fish after chronic parental exposure to an antibiotic. The results showed that the antibiotic could be transmitted to multiple generations and weaken the immune defense of offspring.

Summary: Natural products are considered an important resource for discovering new drugs or potential compounds. This study synthesized derivatives of sinomenine and tested their anti-tumor activity against various cancer cell lines. The results showed that some of the derivatives had broad-spectrum anti-tumor activity and one of them exhibited stronger binding ability than sinomenine. These findings suggest that these derivatives may have potential for treating breast cancer.

Summary: IL-6 is involved in various physiological processes including immune responses and inflammation. It plays a crucial role in chronic inflammation and cytokine storms, such as seen in COVID-19. The IL-6 amplifier, involving NF-kappa B and STAT3, is a key mechanism in promoting uncontrolled inflammatory responses.

Summary: UHMK1 is frequently upregulated in colorectal cancer and its high expression predicts poor prognosis. Knockdown of UHMK1 inhibits CRC cell proliferation and increases sensitivity to oxaliplatin, while overexpression of UHMK1 promotes CRC cell growth and oxaliplatin resistance. UHMK1 interacts with STAT3 and enhances its activity in regulating gene transcription, thus affecting IL6/STAT3 signaling.

Summary: This study aimed to improve the water solubility and stability of cannabidiol (CBD) using micelle technology. CBD-loaded mixed micelles (CBD-M) composed of Poloxamer 407 (P407) and rubusoside (RUB) were successfully prepared and showed significant improvement in solubility and encapsulation efficiency. The CBD-M solution remained stable and maintained the antioxidant activity of CBD after dilution and centrifugation. These findings suggest that CBD-M could serve as a promising formulation for enhanced CBD delivery.
