Metformin against Cancer Stem Cells through the Modulation of Energy Metabolism: Special Considerations on Ovarian Cancer

Ovarian cancer is the most lethal gynecologic malignancy among women worldwide and is presumed to result from the presence of ovarian cancer stem cells. To overcome the limitation of current anticancer agents, another anticancer strategy is necessary to effectively target cancer stem cells in ovarian cancer. In many types of malignancies, including ovarian cancer, metformin, one of the most popular antidiabetic drugs, has been demonstrated to exhibit chemopreventive and anticancer efficacy with respect to incidence and overall survival rates. Thus, the metabolic reprogramming of cancer and cancer stem cells driven by genetic alterations during carcinogenesis and cancer progression could be therapeutically targeted. In this review, the potential efficacy and anticancer mechanisms of metformin against ovarian cancer stem cells will be discussed.

Summary: The study developed a new clinical adhesion score (CLAS) to measure the overall clinical morbidity of adhesion-related complications in abdominal and pelvic surgery. Through Delphi study and pilot study, the CLAS includes 22 outcomes and 23 weight factors, correcting the likelihood of symptoms being related to adhesions through scoring.

A multicentre, randomised, open-label, parallel-group Phase 2b study of belotecan versus topotecan for recurrent ovarian cancer

Summary: This Phase 2b study compared the efficacy and toxicity of belotecan and topotecan in recurrent ovarian cancer, showing that belotecan was not inferior to topotecan in terms of overall response rate. Additionally, belotecan was associated with improved overall survival compared with topotecan, especially in patients with PRROC and non-HGSC. Toxicity did not differ significantly between the two treatment groups.

Summary: The study aimed to develop machine learning models predicting platinum sensitivity in patients with high-grade serous ovarian carcinoma (HGSOC). Six variables associated with platinum sensitivity were selected using clinical data from multiple hospitals, and the LR-based model was found to perform the best. A web-based nomogram was developed for clinical use based on the LR model results.

Summary: Through metabolomics analysis, we identified five distinct metabolic groups in HGSOC cells, with most cancer cells showing an increase in metabolites associated with aerobic glycolysis and phospholipid metabolism. Additionally, subsets of HGSOC cells exhibited elevated levels of 5-Hydroxytryptamine, gamma-aminobutyrate, or glutamate. These findings suggest these metabolites could serve as oncometabolites and potential therapeutic targets in ovarian cancer.

Summary: This study reveals the crucial role of N-linked glycosylation in the functional post-translational modification of KIAA1324, which is important for suppressing cancer cell proliferation and migration in gastric cancer. Inhibition of glycosylation and fucosylation also inhibits KIAA1324-induced cell growth inhibition and apoptosis. Additionally, the study shows that N-linked glycosylation decreases protein stability through rapid proteasomal degradation. Overall, this research highlights the significance of post-translational modifications in KIAA1324 for gastric cancer progression.
