Summary: The RNA polymerase III (Pol III) transcriptome consists of short, highly structured noncoding RNA (ncRNA) and plays important roles in various cellular activities. Recently evolved ncRNAs have expanded the functional repertoire of Pol III transcription in autophagy regulation, immune signaling cascades, and translation. Dysregulation and dysfunction of Pol III transcriptome and ncRNAs are frequently observed in cancer, indicating their involvement in cancer progression and survival outcomes. This review provides an overview of the features, functions, and dysregulation of Pol III transcriptome and ncRNAs in cancer.
