Tyrosine Sulfation of the Amino Terminus of PSGL-1 Is Critical for Enterovirus 71 Infection

Enterovirus 71 (EV71) is one of the major causative agents of hand, foot, and mouth disease, a common febrile disease in children; however, EV71 has been also associated with various neurological diseases including fatal cases in large EV71 outbreaks particularly in the Asia Pacific region. Recently we identified human P-selectin glycoprotein ligand-1 (PSGL-1) as a cellular receptor for entry and replication of EV71 in leukocytes. PSGL-1 is a sialomucin expressed on the surface of leukocytes, serves as a high affinity counterreceptor for selectins, and mediates leukocyte rolling on the endothelium. The PSGL-1-P-selectin interaction requires sulfation of at least one of three clustered tyrosines and an adjacent O-glycan expressing sialyl Lewis x in an N-terminal region of PSGL-1. To elucidate the molecular basis of the PSGL-1-EV71 interaction, we generated a series of PSGL-1 mutants and identified the post-translational modifications that are critical for binding of PSGL-1 to EV71. We expressed the PSGL-1 mutants in 293T cells and the transfected cells were assayed for their abilities to bind to EV71 by flow cytometry. We found that O-glycosylation on T57, which is critical for PSGL-1-selectin interaction, is not necessary for PSGL-1 binding to EV71. On the other hand, site-directed mutagenesis at one or more potential tyrosine sulfation sites in the N-terminal region of PSGL-1 significantly impaired PSGL-1 binding to EV71. Furthermore, an inhibitor of sulfation, sodium chlorate, blocked the PSGL-1-EV71 interaction and inhibited PSGL-1-mediated viral replication of EV71 in Jurkat T cells in a dose-dependent manner. Thus, the results presented in this study reveal that tyrosine sulfation, but not O-glycosylation, in the N-terminal region of PSGL-1 may facilitate virus entry and replication of EV71 in leukocytes.

Summary: Noroviruses are found to be prevalent among hospitalized children in Indonesia, particularly in those under 2 years old. Different predominant genotypes were observed in different years, with GII.4 Sydney_2012 being the most common genotype from 2015 to 2018. The study highlights the significance of continuous molecular surveillance of emerging norovirus strains.

Clinical symptoms observed in these patients included diarrhea, vomiting, fever, cough, rhinorrhea, and dehydration.

The Molecular Evolution of Type 2 Vaccine-Derived Polioviruses in Individuals with Primary Immunodeficiency Diseases

Summary: The use of oral poliovirus vaccine (OPV) has been crucial for preventing person-to-person transmission of poliovirus, but challenges such as cVDPV2 and iVDPV infections pose risks for global polio eradication efforts. The identification of chronic type 2 iVDPV (iVDPV2) excretors highlights the potential risk of inapparent iVDPV infection for expanding cVDPV outbreaks. Further research on genetic characterizations and molecular evolution of iVDPV2 will be critical for elucidating the remaining risk of iVDPV2 in the post-OPV era.

A fatal case of acute encephalopathy in a child due to coxsackievirus A2 infection: a case report

Summary: This study presents a rare case of acute encephalopathy caused by coxsackievirus A2 (CV-A2) in a previously healthy female child. The patient's condition rapidly deteriorated within 2 hours, leading to death. This is the first report of a fatal acute encephalitis-dependent on CV-A2 infection in a child.

Immunogenicity of Catch-Up Immunization with Conventional Inactivated Polio Vaccine among Japanese Adults

Outbreaks of Circulating Vaccine-Derived Poliovirus in the World Health Organization Western Pacific Region, 2000-2021

Summary: Despite the World Health Organization Western Pacific Region (WPR) maintaining a polio-free status for over two decades, the risk of polio outbreaks associated with circulating vaccine-derived polioviruses (cVDPVs) cannot be ignored. Previous cVDPV outbreaks have been effectively controlled, but the frequency and magnitude of such outbreaks in the WPR have been increasing. Therefore, it is important to reconsider the risks of future polio outbreaks associated with cVDPVs and update immunization and surveillance strategies accordingly.
