Local Function Conservation in Sequence and Structure Space

We assess the variability of protein function in protein sequence and structure space. Various regions in this space exhibit considerable difference in the local conservation of molecular function. We analyze and capture local function conservation by means of logistic curves. Based on this analysis, we propose a method for predicting molecular function of a query protein with known structure but unknown function. The prediction method is rigorously assessed and compared with a previously published function predictor. Furthermore, we apply the method to 500 functionally unannotated PDB structures and discuss selected examples. The proposed approach provides a simple yet consistent statistical model for the complex relations between protein sequence, structure, and function. Summary: Males exhibit greater variability than females in many traits, which may have implications for understanding sex differences in health and disease. A mega-analysis of brain structure variability based on MRI data of 16,683 healthy individuals spanning nine decades of life reveals significant patterns of greater male than female between-subject variance in subcortical volumes, cortical surface area, and cortical thickness. These sex differences are present in childhood and may be influenced by early life genetic or gene-environment interaction mechanisms. The findings underscore the importance of individual differences within each sex, which may contribute to sex-specific vulnerability to disorders.

Summary: This study investigated the relationship between childhood trauma severity and gray matter volume. The findings suggest that childhood trauma is associated with reduced gray matter in the frontal lobe, regardless of psychiatric diagnoses. Acoustic speech markers for schizophrenia-spectrum disorders: a diagnostic and symptom-recognition tool

Summary: This study assessed the diagnostic value of acoustic speech parameters in schizophrenia-spectrum disorders and found that automatically extracted speech parameters can accurately classify patients and controls, as well as differentiate between different symptom subtypes. Negative content in auditory verbal hallucinations: a natural language processing approach

Summary: Neuroanatomical abnormalities have been reported to be present from high schizotypy to chronic psychosis. A study conducted a large-scale meta-analysis of cortical and subcortical morphometric patterns of schizotypy in healthy individuals, revealing thicker right medial orbitofrontal/ventromedial prefrontal cortex associated with higher schizotypy scores. The cortical thickness profile in schizotypy showed positive correlation with abnormalities in schizophrenia, while the subcortical volume pattern demonstrated negative correlation with abnormalities in schizophrenia, bipolar disorder, and major depression.

Antipsychotic medication for women with schizophrenia spectrum disorders

Summary: There are significant differences between men and women in the efficacy and tolerability of antipsychotic drugs. Women have slower drug absorption, metabolism, and excretion, leading to higher plasma levels and increased risk for side-effects. Current treatment guidelines are based on studies predominantly conducted in men, which may result in overmedication of women. Women also have higher dopamine receptor occupancy compared to men, necessitating dose adjustments at different hormonal phases.

Summary: Negative emotional valence of auditory verbal hallucinations in schizophrenia is associated with reduced glutamate levels in multiple brain regions, indicating aberrant glutamatergic signaling. Specifically, voices that evoke emotional resistance rather than behavioral resistance are predictive of lower glutamate levels, supporting the glutamate hypothesis of schizophrenia.

Brain vasculature disturbance in schizophrenia

Summary: The review discusses the vascular hypothesis of schizophrenia and provides recent evidence for and against this hypothesis. Increased blood-brain barrier permeability is observed in a consistent proportion of schizophrenia patients and is associated with inflammation and cognitive deficiencies. Gender also interacts with blood-brain barrier integrity and permeability in schizophrenia. The strongest independent genetic association with schizophrenia has been identified in the FZD1 gene. There is evidence of alterations in brain perfusion in schizophrenia, although medication may play a role. The review encourages further investigation into the dynamic context of gene expression and performance of human brain endothelium in relation to schizophrenia.

Role of the gut microbiome in three major psychiatric disorders

Summary: Major depressive disorder (MDD), bipolar disorder (BD), and schizophrenia-spectrum disorders (SSD) are heterogeneous psychiatric disorders that have a significant impact on the well-being and global health of patients. Disruptions in the gut microbiome may contribute to the development of these disorders. Current research suggests that patients with psychiatric disorders tend to have higher levels of Streptococcus, Lactobacillus, and Eggerthella bacteria, and lower levels of Faecalibacterium, a bacteria that produces butyrate. However, further studies are needed to account for confounding factors such as medication use and lifestyle choices, and the effectiveness of probiotics and fecal microbiota transplantation (FMT) in treating these disorders remains inconclusive.

A data-driven linguistic characterization of hallucinated voices in clinical and non-clinical voice-hearers
