Summary: Under the pandemic situation, there is a need for sufficient prophylactic vaccines. Intradermal (ID) vaccination is an effective method, but currently not widely used due to technical difficulties. This study presents the design and performance testing results of the ImmuciseTM Intradermal Injection System. The study shows that ImmuciseTM can inject drugs precisely into the ID tissues and is safe to use. An analysis comparing ID administration with standard subcutaneous (SC) administration of influenza vaccine shows that ID administration is equally or more effective up to 180 days later. These findings suggest that ImmuciseTM is an efficient device for distributing a small amount of vaccine widely during a pandemic.

Summary: The study found that intradermal administration of a live-attenuated vaccinia-based SHIV vaccine provided protection in female macaques, while intramuscular administration did not. Both routes of vaccination induced similar levels of serum IgG with neutralizing and non-neutralizing activities. Intradermal vaccination induced greater germinal center Tfh and B cell responses, reduced the ratio of Th1 to Tfh cells in blood, and showed lower activation of monocytes and inflammasome compared to intramuscular vaccination.

Summary: The multimicrochannel microneedle microporation (4M) platform proposed in this work achieves high efficiency, safety, and uniformity for in vivo intracellular delivery. It utilizes internal microchannels implemented through the microneedle to provide a concentrated, safe electric field that accelerates cargo movement under electrophoresis and triggers cell electroporation for enhanced transport across cell membranes. This platform has shown significant improvements in anticancer effects and reduced systemic toxicity, making it a general-purpose delivery tool for emerging drugs in vivo.

Summary: In this study, a new pulse generator, applicator, and pulse delivery protocol for gene electrotransfer (GET) to skin cells were developed. The results showed that the proposed pulse delivery protocol resulted in higher gene expression in mouse skin cells compared to the currently used pulse generator, applicator, and pulse delivery protocol.

Summary: In this study, microfluidics was used to perform CRISPR/Cas9 gene editing and transduction, resulting in the generation of CAR T cells targeting B cell antigen. Through device optimization and comprehensive testing, high editing efficiency and cell viability were achieved. Microfluidics-based manufacturing showed higher yield and similar cell viability and functionality compared to electroporation.

Summary: An estimated 350 million people worldwide are living with chronic Hepatitis B virus, with preventative HBV vaccination in infants reducing disease burden to some extent. However, vulnerable populations in endemic regions are still at risk of infection. Gene electrotransfer (GET) combined with moderate heating can achieve higher levels of gene expression than GET alone, potentially serving as a strong immune response inducing platform.

Summary: Electrotransfection (ET) is a non-viral approach used for delivering nucleic acids, but its efficiency is often low in vivo. In this study, we developed nanoenhancers that diffuse slowly in tissues and are poorly absorbed by blood and lymph microvessels. These nanoenhancers were found to enhance ET efficiency both in vitro and in vivo, and their mechanism of action was related to the inhibition of lysosomal functions.
