Summary: The exposure to AAI/AAII mixture may enhance AA-DNA adduct formation in rat livers and kidneys, partially due to inhibition of AAI detoxification and induction of NQO1. Additionally, AAII may act as an inhibitor of AAI detoxification, leading to increased AAI residues in liver and kidney tissues and enhanced AAI DNA adduct formation.

Summary: This study investigated the potential role of Apurinic/apyrimidine endonuclease 1 (APE1) in aristolochic acid I (AAI)-induced nephrotoxicity. The results showed that AAI inhibited the expression of APE1, leading to oxidative damage and apoptosis. Inhibition of APE1 further decreased the expression of Nrf2 and HO-1 proteins, and aggravated AAI-induced apoptosis. In contrast, overexpression of APE1 promoted the expression of Nrf2 and HO-1, and alleviated AAI-induced apoptosis and renal injury. Targeting APE1 could be an effective therapeutic strategy for treating AAI nephrotoxicity.
